ABSTRACT Testicular ageing is accompanied by a series of morphological changes, while the features mitochondrial dysfunction remain largely unknown. Herein, we observed range age‐related modifications in testicular morphology and spermatogenic cells, conducted single‐cell RNA sequencing on young old testes Drosophila . Pseudotime trajectory revealed significant changes germline subpopulations during ageing. Our examination unveiled that genes showing bias spermatids exhibited higher dN/dS than those GSCs_Spermatogonia. Genes biased towards GSCs_Spermatogonia displayed Interestingly, demonstrated lower contrast to spermatids, revealing complexity evolutionary adaptations Furthermore, mitochondria associated events, including oxidative phosphorylation, TCA cycle pyruvate metabolism, were significantly enriched subpopulations. Specifically, function was impaired process ageing, concurrently emphasising role several key nuclear genome‐encoded regulatory genes, such as Hsp60B, fzo, Tim17b1 mRpL12. data offer insights into homeostasis regulated process.
