Rheumatoid arthritis (RA) has been associated with a relative expansion of faecal Prevotellaceae. To determine the microbiome composition and prevalence Prevotella spp. in group individuals at increased risk for RA, but prior to development disease. In an ongoing cohort study first-degree relatives (FDRs) patients we identified 'FDR controls', asymptomatic without autoantibodies, pre-clinical RA stages, who had either developed anticitrullinated peptide antibodies or rheumatoid factor positivity and/or symptoms signs possible RA. Stool sampling culture-independent microbiota analyses were performed followed by descriptive statistics statistical community structures.A total 133 participants included, which 50 categorised as controls' 83 'pre-clinical stages'. The stages' was significantly altered compared FDR controls. We found significant enrichment bacterial family Prevotellaceae, particularly spp., RA' (p=0.04).Prevotella stages before onset suggests role intestinal dysbiosis

Abstract The trillions of microorganisms in the gut microbiome have attracted much attention recently owing to their sophisticated and widespread impacts on numerous aspects host pathophysiology. Remarkable progress large-scale sequencing mass spectrometry has increased our understanding influence and/or its metabolites onset progression extraintestinal cancers efficacy cancer immunotherapy. Given plasticity microbial composition function, microbial-based therapeutic interventions, including dietary modulation, prebiotics, probiotics, as well fecal transplantation, potentially permit development novel strategies for therapy improve clinical outcomes. Herein, we summarize latest evidence involvement immunity metabolism, effects immune response, modulate microbiome, discuss ongoing studies future areas research that deserve focused efforts.

Microfold (M) cells are located in the epithelium covering mucosa-associated lymphoid tissues, such as Peyer's patches (PPs) of small intestine. M actively transport luminal antigens to underlying follicles initiate an immune response. The molecular machinery M-cell differentiation and function has been vigorously investigated over last decade. Studies have shed light on role mucosal system revealed that antigen uptake by contributes not only but also systemic responses. However, studies usually focus infectious diseases; contribution autoimmune diseases remained largely unexplored. Accumulating evidence suggests dysbiosis intestinal microbiota is implicated multiple diseases, including diseases. This implies microorganisms PPs may play a pathogenesis In this review, we relationship between commensal bacteria We provide outline current understanding biology subsequently discuss potential induction autoimmunity, beyond

The barrier surfaces of the gastrointestinal tract are in constant contact with various microorganisms. Cytokines orchestrate mucosal adaptive and innate immune cells defense against pathogens. IL-10 IL-22 most well studied members family play essential roles maintaining homeostasis. serves as an important regulator preventing pro-inflammatory responses while plays a protective role tissue damage contributes to pathology certain settings. In this review, we focus on these two cytokines development diseases, including inflammatory bowel diseases (IBD) colitis-associated cancer (CAC). We summarize recent studies try gain better understanding how they regulate maintain equilibrium under conditions.

Human health and disease have increasingly been shown to be impacted by the gut microbiota, mouse models are essential for investigating these effects. However, compositions of human microbiotas distinct, limiting translation microbiota research between hosts. To address this, we constructed Mouse Gastrointestinal Bacteria Catalogue (MGBC), a repository 26,640 high-quality microbiota-derived bacterial genomes. This catalog enables species-level analyses mapping functions interest identifying functionally equivalent taxa humans mice. We complemented this with publicly deposited collection 223 isolates, including 62 previously uncultured species, facilitate experimental investigation individual commensal bacteria in vitro vivo. Together, resources provide ability identify test members host-specific mice support informed use research.
