Background: Recent studies suggest that the anterior limb of internal capsule may be an area convergence for multiple compulsion loops. In this study, role different dopaminergic loops in mechanism obsessive-compulsive disorder (OCD) was investigated by selectively damaging neurons or fibers corresponding targets with 6-hydroxydopamine (6-OHDA) and depicting anatomical map various located capsule. Methods: A total 52 male Sprague Dawley (SD) rats were exposed to either saline (1 mL/kg, NS group, n = 6) quinpirole (QNP, dopamine D2-agonist, 0.5 mg/kg, 46) twice weekly 5 weeks. After each injection, placed on open field analyze aspects their behaviour, including number home base visits (NOH), average time between visit (ATBO), distance travelled (TDM). model setup, 46 QNP divided randomly into five groups: 6-OHDA (AC) stereotactic injection group (QNP+AC 10), mediodorsal thalamic nucleus (MD) (QNP+MD accumbens (NAC) (QNP+NAC (QNP+NS-S non-surgical (QNP+Non-S 6). simultaneously received a (NS+6-OHDA-S 6: AC2, MD2, NAC2). All QNP-treated then continued given week 4 weeks, behaviour observed after infusion. immunofluorescence staining used monitor distribution nerve areas intervention loops, quantitative analysis performed. Results: Compulsive declined gradually QNP+AC QNP+NAC groups 3 weeks surgery, decreasing more rapidly. The QNP+MD had decreased surgery but increased almost same level as pre-surgery at post-surgery. Postoperative fluorescence suggested (DA) target significantly reduced compared QNP+Non-S group. Conclusions: damages within OCD loop, which somewhat alleviates compulsive behaviours. results AC might best therapeutic interventions OCD.

Background Constipation is a common digestive system disorder, which closely related to the intestinal flora. Zhishi Daozhi decoction (ZDD) traditional Chinese medicine prescription used treat constipation caused by indigestion. This study evaluate efficacy of ZDD in treating and elucidate underlying mechanism. Methods In this study, Kunming mice were administered high-protein diet (HFHPD) loperamide hydrochloride injections induce constipation. The then received varying doses (2.4, 4.7, 9.4 mg/kg) for seven days. Following sampling process, we measured fecal microbial activity. levels 5-hydroxytryptamine (5-HT), vasoactive peptide (VIP), aquaporin-3 (AQP3) quantified using enzyme-linked immunosorbent assay. Changes gut microbiota evaluated through 16S rRNA gene sequencing. Additionally, investigated correlation between specific features 5-HT, VIP, AQP3. Results surface model group (CMM) was rough dry. stool low-dose (CLD), medium-dose (CMD), high-dose (CHD) exhibited smoother texture, resembling that normal (CNM). 5-HT CMM significantly lower than CNM, CLD, CHD. VIP CMD other four groups, AQP3 showed decreasing trend. activity higher groups. Diversity analysis indicated CHD treatments more effective restoring structure. Potential pathogenic bacteria, including Clostridium , Aerococcus Jeotgalicoccus Staphylococcus enriched CMM. contrast, beneficial bacteria such as Faecalibacterium Bacillaceae Bacillus prevalent CMD, Correlation revealed Streptococcus Enterococcus positively correlated with while Succinivibrio negative 5-HT. Conclusions induced HFHPD disrupts structure microbiota. appears alleviate constipation, potentially mechanisms linked brain-gut axis its interaction Among treatment medium dose demonstrated most results.

Abstract Neuroinflammation, a key defense mechanism of the nervous system, is associated with changes in inflammatory markers and stimulation neuroimmune cells such as microglia astrocytes. Growing evidence indicates that gut microbiota its metabolites directly or indirectly regulate host health. According to recent studies, bacterial dysbiosis closely linked several central system disorders cause neuroinflammation, including multiple sclerosis, Alzheimer's disease, Parkinson's sepsis‐associated encephalopathy, ischemic stroke. Recent findings indicate bidirectional communication network between influences neuroinflammation cognitive function. Dysregulation this can affect generation cytotoxic metabolites, promote impair cognition. This review explores lesser‐studied microbiota‐derived involved neuroinflammation—bile acids, trimethylamine‐N‐oxide, indole derivatives—as targets for creating new treatment tools neuroinflammatory illnesses, well possible biomarkers early diagnosis prognosis.

The neurobehavioral changes in food allergy mice have not been comprehensively studied, and the mechanism underlying them remains unclear. Our study aims to fully investigate OVA (ovalbumin)-sensitized explore potential via gut microbiota–brain axis. We established mouse (C57BL/6J male) model with OVA, evaluating anaphylactic symptoms levels of Th2 signature cytokine allergy-related antibodies serum. Using behavioral tests, we measured anxiety, depression, social behavior, repetitive attention, spatial memory control mice. In addition, analyzed prefrontal cortex for measuring inflammation-related indicators gathered serum untargeted metabolomics analysis feces 16S rDNA sequencing. exhibited significantly elevated IgE levels. addition anxiety-like, depression-like, behaviors, also displayed less interest damaged attention. TNF-α, IL-1β, IL-6 activation microglia were increased, which might explain neuronal damage. multi-omics technology, amino acid metabolism disruption, particularly carboxylic acids derivatives, was observed mice, remarkably correlated altered abundance microbiota related allergy. Behaviors OVA-induced extensively impaired. disruption associated dysbiosis play a pivotal role impairing neural immune homeostasis damage, could be responsible abnormalities.

A growing body of literature suggests a link between bowel syndromes (e.g., irritable syndrome and inflammatory disease), gut microbiome alterations, psychiatric disorders. This narrative review aims to explore the potential role in pathogenesis clinical presentation obsessive–compulsive disorder (OCD) whether there is sufficient evidence warrant considering gastrointestinal symptoms their implication for during assessment treatment OCD. For this purpose, PubMed search studies focusing on OCD, microbiota, syndrome, disease was conducted by two independent reviewers. While current issues OCD remains limited, emerging alterations high rates population. These findings emphasize importance incorporating comprehensive assessments into “global OCD”. Such should encompass various factors, including physical comorbidities symptoms, nutritional habits, fluid intake, exercise patterns, dysfunctions inflammation. Considering implications, interventions targeting health, such as probiotics dietary modifications, may hold promise improving patients with comorbid problems. Further research area warranted better understand interplay health effectiveness targeted outcomes.

Escherichia coli Nissle 1917 (EcN), the probiotic featured with well-established safety in different host, is emerging as a favored chassis for construction of engineered probiotics disease treatment. However, limited by low intestinal colonization ability EcN, repeated administration required to maximize health benefits EcN-derived probiotics. Here, using fecal metabolites "metabolites pool", we developed metabolomic strategy characterize comprehensive metabolic profile EcN. Compared Prevotella copri DSM 18205 (P. copri), one dominant microbes gut flora, EcN exhibited minor growth advantage under metabolites-containing condition its lower capability towards metabolites. Further study indicated that lacked import oligopeptides containing more than two amino acids. The shortage oligopeptides-derived acids might limit restricting purine metabolism. Assisted bioinformatic and qRT-PCR analyses, identified tripeptides-specific importer Pc-OPT P. copri, which was mainly distributed genera Bacteroides. Overexpression improved tripeptides importation promoted colonization. Notably, 16S rRNA gene amplicon sequencing results strengthening promote adjusting microbial composition. Our reveals insufficient paves road promoting efficacy synthetic improving their ability.
