Human papillomavirus (HPV)16-E6 seropositivity accurately predicts oropharyngeal squamous cell carcinoma (OPSCC) risk decades before diagnosis; but the biomarker's translational potential is unknown. To inform considerations for OPSCC screening, we described HPV16-E6 seroprevalence, predictors, and kinetics among cancer-free men.

In 80% young women, there clearance within 18–24 months. developed countries, oral squamous cell carcinoma (OSCC) now frequent human (HPV)-related cancer, having surpassed cervical and it predicted by 2030 carcinomas will be HPV-related rather non-HPV-related form. However, are currently no screening programs for cavity infection. While natural history HPV cervix well known, oropharynx, not entirely clear. Furthermore, prevalence oropharynx unknown. Published studies have found wide-ranging estimates 2.6% to 50%. There also conflicting results regarding percentage presenting same type at two mucosal sites, ranging from 0 60%. Additionally, question arises as whether can develop infection, through contact or self-inoculation, should considered an independent event. still consensus on these topics, nor relationship between infections. Published: Feb. 26, 2025

The strong association between human papillomavirus (HPV) infection and oropharyngeal squamous cell carcinoma (SCC) is well-documented, with p16 expression serving as a reliable predictor of HPV involvement. HPV-related tumors are characterized by distinct mechanisms affecting p53 protein pathways. However, the prevalence combined predictive utility in head neck cutaneous SCC remain less explored, necessitating further research to understand their role this subset SCC. HPV, p16, statuses were determined using immunohistochemistry staining methods rather than more sensitive techniques such polymerase chain reaction or genotyping, limiting ability assess specific area types potentially associated prognosis. Further studies assessing multiple molecular markers patients will better predict diverse outcomes type treatment targeted those markers.

Introduction Tumor-associated macrophages (TAMs) recruited from circulating monocytes drive tumor-growth and establish an immunosuppressive tumor microenvironment (TME). Initial events in transition resting to TAMs are poorly understood. Here, we report that oropharyngeal cancer (OPC) patients control treated with OPC-conditioned media (CM) express a repertoire of pro-tumor mediators is characteristic TAMs. Methods Monocytes were stimulated OPC cell line CM, analyzed by single-cell RNAseq. Results select genes confirmed qPCR tumors vs. clinically normal tissue. spheroids containing T-cells established, TAM phenotype characterized flow analysis qPCR, T-cell proliferation assessed flow. induced multiple including CXCL1, CXCL5, CXCL8, SPP1, IL1B, GPNMB , FABP5 . Patient had higher baseline levels or achieved after stimulation than monocytes. A subset patient high CXCL9/-10/-11 expression resisted downregulation response stimulation, potential sign more favorable TME. biopsies compared tissue correlated outcome. Spheroid derived maintained the seen conditioned media. These suppress proliferation. Inhibition COX-2 IL1 signaling during differentiation into partially blocked suppression Conclusion Targeting early could be used develop new therapies for OPC.
