Abstract This study was conducted to assess the effect of Evodiae Fructus 70% ethanol extract (EFE) on pathology atopic dermatitis using in vitro and vivo models. The major compounds EFE were identified by ultra-performance liquid chromatography with tandem mass spectrometry as rutaecarpine, evodiamine, evodol, dehydroevodiamine, limonin, synephrine, evocarpine, dihydroevocarpine, hydroxyevodiamine. significantly decreased chemokine levels tumor necrosis factor-α/interferon-γ-stimulated HaCaT cells. In house dust mite-treated NC/Nga mice, topical application score, epidermal hyperplasia thickening, mast cell infiltration, plasma histamine corticosterone. Thymic stromal lymphopoietin, CD4 + T cells, interleukin-4, intercellular adhesion molecule-1 expression lesioned skin reduced treated mice. mechanism elucidated transcriptome analysis, followed experimental validation Western blotting down-regulated activation Janus kinase (JAK)-signal transducers activators transcription (STAT) mitogen-activated protein kinases (MAPK) signaling pathways improves dermatitis-like symptoms suppressing inflammatory mediators, cytokines, chemokines regulating JAK-STAT MAPK pathways, suggesting its use a potential agent for treatment dermatitis.

Rheumatoid arthritis (RA) is a highly prevalent autoimmune disorder. The pathogenesis of the disease complex and involves various cellular populations, including fibroblast-like synoviocytes, macrophages, T cells, among others. Identification signalling pathways molecules that actively contribute to development crucial understanding mechanisms involved in chronic inflammatory environment present affected joints. Recent studies have demonstrated Janus kinase/signal transducer activator transcription (JAK/STAT) pathway regulates behaviour immune cells contributes progression RA. Several JAK inhibitors, such as tofacitinib, baricitinib, upadacitinib, filgocitinib, been developed, their efficacy safety patients with RA comprehensively investigated number clinical trials. Consequently, inhibitors approved registered treatment for In this review, we discuss involvement JAK/STAT summarise potential beneficial effects implicated disease. Moreover, most important phase 3 trials evaluated use these agents patients.

The intratumoral microbiome (TM) refers to the microorganisms in tumor tissues, including bacteria, fungi, viruses, and so on, is distinct from gut circulating microbiota. TM strongly associated with tumorigenesis, progression, metastasis, response therapy. This paper highlights current status of TM. Tract sources, adjacent normal tissue, circulatory system, concomitant co-metastasis are main origin advanced techniques analysis comprehensively summarized. Besides, involved progression through several mechanisms, DNA damage, activation oncogenic signaling pathways (phosphoinositide 3-kinase [PI3K], signal transducer activator transcription [STAT], WNT/β-catenin, extracellular regulated protein kinases [ERK]), influence cytokines induce inflammatory responses, interaction microenvironment (anti-tumor immunity, pro-tumor microbial-derived metabolites). Moreover, promising directions therapy include immunotherapy, chemotherapy, radiotherapy, application probiotics/prebiotics/synbiotics, fecal transplantation, engineered microbiota, phage therapy, oncolytic virus inherent challenges clinical also review provides a comprehensive landscape for analyzing TM, especially TM-related mechanisms TM-based treatment cancer.

Disturbances in immunoregulation may lead to both cancer and autoimmune diseases. Many therapeutic drugs for diseases also display anti-tumor efficacy. The Janus kinase/signal transducer activator of transcription signaling pathways are involved the secretion more than 50 distinct cytokines, which have critical roles inducing tumorigenesis. Thus, kinases become classical immunotherapeutic targets immune disease. More 70 kinase inhibitors been approved as immunomodulatory clinical use, 12 used treatment This systematic review aims elucidate role clinically that were primarily designed their potential translation treatments.
