The incidence of pituitary adrenocorticotropic hormone (ACTH)-secreting PitNETs, commonly known as ACTH is significantly higher in females; however, the underlying causes for this gender disparity remain unclear. In study, we analyzed expression deubiquitinating enzymes functioning PitNETs from both male and female subjects using RNA sequencing identified USP11 a potential susceptibility factor contributing to prevalence these females. Further investigation revealed that markedly elevated with levels than those observed normal tissue. Experimental data indicate promotes transcription proopiomelanocortin (POMC) secretion ACTH. contrast, knockdown leads substantial reduction POMC secretion, demonstrated vitro vivo models. Mechanistically, found facilitates deubiquitination key TPIT enhancing its protein stability thereby promoting secretion. Additionally, virtual screening Lomitapide Nicergoline inhibitors USP11, reducing Thus, emerges therapeutic target, drugs aimed at inhibiting function could benefit women Cushing's disease.

Apatinib, a commonly used tyrosine kinase inhibitor in cancer treatment, can cause adverse reactions such as hypertension. Hypertension, turn, increase the risk of certain cancers. The coexistence these diseases makes use combination drugs more common clinical practice, but potential interactions and regulatory mechanisms drug combinations are poorly understood. We humanized CYP2D6 mouse model to predict effect apatinib on pharmacokinetics pharmacodynamics metoprolol investigated interactional mechanism. inhibitory effects were vitro by using wild-type liver microsomes (WT MLMs), (hCYP2D6 human (HLMs). Molecular docking was utilized explore structural basis observed mode. And vivo interaction between assessed study mice. In studies molecular experiments indicated that competitively inhibited metabolism metoprolol. findings revealed administration combined with resulted significant AUC(0-t), AUC(0-∞) Cmax metoprolol; additionally, there reduction CLz/F heart rate, indicating strongly metabolism. homologous protein WT mice sensitive compared hCYP2D6 Gender analysis accumulation pronounced male when apatinib, higher susceptibility cardiotoxicity males.

Abstract Background Psoriasis is a prevalent multifactorial chronic inflammatory skin disease that may adversely impact kidney function in affected individuals. This study aimed to investigate estimated glomerular filtration rate (eGFR) hospitalized patients with psoriasis, analyze its influencing factors, and explore the effects of sex age on eGFR psoriasis. Methods retrospectively analyzed values 456 psoriasis who were admitted two hospitals Taizhou, Zhejiang Province from January 2012 December 2023 using multiple linear regression. Results The prevalence (CKD) among was 7.9% (36/456). Multiple regression analysis revealed (B = -5.50, P 0.014), -23.10, < 0.001), duration -4.36, 0.018), hypertension -9.41, cardiovascular -8.68, 0.019), gout -18.73, 0.001) as main factors Subgroup by -22.32, -4.95, 0.021), -10.65, -17.56, male whereas -24.75, -32.83, 0.002), -44.00, 0.015) female -6.63, 0.015), -6.51, 0.006), -10.90, -14.84, 0.011) aged 60 years, -8.08, 0.008), -11.21, 0.020), -26.41, 0.005) ≥ years. Conclusion research indicated that differences sex. Hence, corresponding measures should be formulated for different ages sexes clinical settings.

Sex differences in metabolic disorders and susceptibility to chronic diseases induced by a high-fat diet (HFD) exhibit significant dimorphic characteristics. A long-standing male-centric bias medical research healthcare, predominantly focused on male physiological traits, has hindered the precise treatment of female patients. comprehensive understanding sex health their underlying mechanisms is crucial for advancing personalized promotion precision medicine. This review systematically elucidates sex-specific manifestations diet-associated disorders: males develop visceral adiposity, insulin resistance, dyslipidemia, accompanied significantly elevated risk cardiovascular syndromes. Premenopausal females maintain homeostasis through estrogen-mediated optimization glucose lipid metabolism oxidative stress buffering mechanisms, whereas postmenopausal-phase experience dramatic vulnerability due z loss protective barriers. Furthermore, we emphasize multidimensional mechanistic interpretations sexual dimorphism from perspectives including chromosome complement, hormone signaling pathways, epigenetic regulation, gut microbiota composition, neuroendocrine dimorphism. work provides critical theoretical foundations rectifying unisex paradigms optimizing early warning systems therapeutic strategies disorders.

Abstract Breast cancer is the leading cause of cancer-related mortality among women worldwide. The development predictive biomarkers and immunologic markers has revolutionized breast diagnosis treatment, enabling personalized medicine approaches. This study aims to review current status, challenges, future directions in cancer. Predictive such as HER2/neu expression, estrogen receptor (ER)/progesterone (PR) genomic signatures play a crucial role treatment decisions, particularly for hormone receptor-positive HER2-positive tumors. Immunologic markers, including PD-L1 microsatellite instability (MSI), tumor mutational burden (TMB), tumor-infiltrating lymphocytes (TILs), are essential predicting responses immunotherapy, especially aggressive subtypes like triple-negative (TNBC). However, challenges heterogeneity, therapy resistance, standardization biomarker testing remain significant barriers. also explores emerging technologies artificial intelligence (AI), profiling, liquid biopsy, which expected enhance precision medicine, improve early diagnosis, optimize strategies. integration these advanced molecular approaches holds great promise improving patient outcomes overcoming existing limitations management.
