Mechanistic understanding of acetoclastic methanogenesis is pivotal for optimizing anaerobic digestion efficient methane production. In this study, two different operational modes, continuous flow reactor (CFR) and sequencing batch (SBR), accompanied with solids retention times (SRT) 10 days (SBR10d CFR10d) 25 (SBR25d CFR25d) were implemented to elucidate their impacts on microbial communities energy metabolism methanogens in acetate-fed systems. Microbial community analysis revealed that the relative abundance Methanosarcina (16.0% ∼ 46.0%) surpassed Methanothrix (3.7% 22.9%) each reactor. SBRs had potential enrich both Methanosarcina. Compared SBRs, CFRs lower total methanogens. exhibited a superior enrichment reactors 10-day SRT, while preferred be acclimated 25-day SRT. ABSTRACT Biogas reactors operating at elevated ammonia levels are commonly susceptible to process disturbances, further augmented thermophilic temperatures. The major cause is assumed be linked inhibition followed by an imbalance between different functional microbial groups, centred around the last two steps of anaerobic digestion, involving acetogens, syntrophic acetate oxidisers (SAOB) and methanogens. Acetogens key contributors reactor efficiency, acting as crucial link hydrolysis fermentation final methanogenesis step. Their product acetate, high converted SAOB hydrogenotrophic methanogens biogas. Even though these functionally processes well recognised, less known about responsible organism temperature conditions. main aim this study was garner insights into penultimate stages in three (52°C) operated under (FAN 0.7–1.0 g/L; TAN 3.6–4.4 g/L). primary objective identify potential acetogens SAOBs. Metagenomic data from were analysed for reductive acetyl‐CoA pathway (Wood–Ljungdahl Pathway) glycine synthase reductase pathway. results revealed a lack true but uncovered candidates that harbour WLP, ‘ Candidatus Thermodarwinisyntropha acetovorans’, Thermosyntrophaceticus schinkii’, Thermotepidanaerobacter aceticum’, lipid‐degrader Thermosyntrophomonas ammoiaca’.

The current focus on renewable energy in global policy highlights the importance of methane production from biomass through anaerobic digestion (AD). To improve while ensuring overall process stability, microbiome-based management strategies become more important. In this study, metagenomes and metaproteomes were used for metagenomically assembled genome (MAG)-centric analyses to investigate a full-scale biogas plant consisting three differentially operated digesters. Microbial communities analyzed regarding their taxonomic composition, functional potential, as well functions expressed proteome level. Different abundances genes enzymes related could be mostly attributed different parameters. Individual MAGs exhibiting digesters studied detail, roles hydrolysis, acidogenesis acetogenesis steps assigned.
