Cardiolipin (CL) plays a critical role in maintaining mitochondrial membrane integrity and overall homeostasis. Recent studies have suggested that damage resulting from abnormal cardiolipin remodelling is associated with the pathogenesis of diabetic kidney disease (DKD). Acyl-coenzyme A:lyso-cardiolipin acyltransferase-1 (ALCAT1) was confirmed to be involved progression Parkinson's disease, diet-induced obesity other ageing-related diseases by regulating pathological remodelling. Thus, purpose this investigation determine ALCAT1-mediated CL DKD explore potential underlying mechanism.

We investigate the mechanism of action astragalin (AST) in treatment Alzheimer's disease (AD). Network pharmacology was conducted to analyze relationships among AST, AD, and neuroinflammation, The APP/PS1 transgenic mice with AD were used experiments; be specific, influence AST on behavior analyzed by Morris water maze eight-arm radial tests, tissue inflammatory factor levels detected ELISA, pathological changes H&E immunohistochemical staining. Analysis results network suggested that exerted multi-target effect neuroinflammation AD. Through molecular docking dynamics analyses, COX2 might target AST. Moreover, animal experimental demonstrated improved mice, enhanced motor memory abilities, meanwhile, it suppressed expression factors tissues activation microglial cells. this study discovers can suppress cell via improve

ABSTRACT Metabolic syndrome (MetS) is a cluster of interrelated metabolic abnormalities that significantly elevate the risk cardiovascular disease, obesity, and diabetes. Flavonoids, diverse class bioactive polyphenolic compounds found in plant‐derived foods beverages, have garnered increasing attention as potential therapeutic agents for improving health. This review provides comprehensive analysis effects flavonoids context MetS, with particular focus on their modulation AMP‐activated protein kinase (AMPK) pathway. AMPK serves central regulator cellular energy balance, glucose metabolism, lipid homeostasis, making it critical target intervention. Through systematic literature up to April 2024, preclinical studies across various flavonoid subclasses, including flavonols, flavan‐3‐ols, were analysed elucidate mechanistic roles regulation. Many suggests enhance glycolipid metabolism by facilitating transporter 4 (GLUT4) translocation activating pathway, thereby glycemic control diabetes models. In obesity‐related studies, demonstrated significant inhibitory synthesis, reduced adipogenesis, attenuated proinflammatory cytokine secretion via activation. These findings show broad addressing MetS its associated disorders. While these insights highlight promising natural health improvement, important note excessive concentrations may disrupt pathways, potentially leading imbalance cytotoxicity. Further clinical trials are essential determine optimal dosing regimens, formulations, long‐term safety efficacy flavonoids. highlights importance interventions targeting comorbidities, offering foundation future translational research.

Anzi Tiaochong Fang (ATF) is a traditional Chinese medicine (TCM) Fangji widely used to treat antiphospholipid syndrome-related recurrent pregnancy loss (APS-RPL). This study aimed identify the quality markers and elucidate mechanisms of ATF in treating APS-RPL. Chemical, network pharmacology, vitro verification were employed ATF. HPLC-MS/MS was quantify compounds. APS-RPL targets identified using databases such as HERB, similarity ensemble approach, PharmMapper, Swiss Target Prediction, Gene Expression Omnibus, Genecards, DisGeNET. GO Reactome analyses conducted KOBAS-i. In performed CCK-8, FDA staining, ELISA. 23 compounds 942 targets, including 132 42 between analysis demonstrated significant enrichment cytokine-mediated signaling pathway, positive regulation angiogenesis, response hypoxia, inflammatory response, platelet degranulation. indicated Immune System, Cytokine Signaling system, by Interleukins, Platelet activation, aggregation, VEGF. Core included VEGFA, ALB, TNF, IL-6, STAT3, with liquiritigenin, nobiletin, ginsenoside Rb1, astragalin markers. experiments that promoted HTR-8/SVneo cell viability, significantly reduced TNF-α IL-1β levels, upregulated IL-6. These findings contribute identification quantification potential molecular ATF, thereby supporting its therapeutic treatment

Purpose To explore the influencing factors of acute kidney injury in elderly patients with diabetic nephropathy and to construct a nomogram model. Methods The research subjects were 680 type 2 admitted our hospital. included from May 2018 August 2023. Patients used as merge group (n=50), without unmerge (n=630) was included. prevalence predisposing analyzed, multivariate logistic regression analyze patients, risk prediction model established based on for verification. Results Analysis found that severe infection main trigger, accounting 40.00%, followed by nephrotoxic antibiotics heart failure. age, urine microalbumin-to-creatinine ratio (ACR), blood urea nitrogen (BUN), uric acid(UA), cystatin C (CysC) levels combined significantly higher than those ( P <0.05), left ventricular ejection fraction (LVEF) epidermal growth factor receptor (eGFR) lower <0.05). Age, ACR, CysC are independent nephropathy, LVEF eGFR protective C-index predicting is 0.768 (95% CI: 0.663-0.806), calibration curve tends ideal curve; threshold >0.18, provides clinical net benefits, benefits predictors. Conclusion establishment CysC, LVEF, has good predictive effect, which can help doctors more accurately assess patient’s condition provide basis formulating personalized treatment plans.
