Background: Hepatocellular carcinoma (HCC) is frequently diagnosed at late stages when curative treatments are no more appliable. Many studies have proved the active role of long non-coding RNAs (lncRNAs) in cancers’ biology; here, functional lncRNA NCK1-AS1 HCC was identified. Methods: Gene expression tumor tissues evaluated by examining online databases and 88 collected samples from our hospital. The interactions miR-22-3p with tyrosyl-tRNA synthetase (YARS) were tested conducting bioinformatics analysis, luciferase report, RNA pulldown experiments. CCK-8, colony formation, flow cytometry, wound healing, transwell experiments used to dissect NCK1-AS1/miR-22-3p/YARS axis HCC.Results: overexpressed cells tissues. Functional assays depicted that depletion hampered malignant character cells. controlled availability miR-22-3p, resulting YARS upregulation. found a clinical value for diagnosis. Moreover, rescue revealed inhibition or overexpression partially blocked function deficiency As downstream signaling pathway, we discovered activated PI3K/AKT miR-22-3p/YARS axis. Conclusion: present study verified could promote progression via activate signaling.

Post-transcriptional modifications to coding and non-coding RNAs are unquestionably a pivotal way in which human mRNA protein diversity can influence the different phases of transcript's life cycle. CELF (CUGBP Elav-like family) proteins RBPs (RNA-binding proteins) with pleiotropic capabilities RNA processing. Their responsibilities extend from alternative splicing transcript editing nucleus stability, translation into cytoplasm. In this way, family members have been connected global alterations cancer proliferation invasion, leading their identification as potential tumor suppressors or even oncogenes. Notably, genetic variants, splicing, phosphorylation, acetylation, subcellular distribution, competition other RBPs, ultimately lncRNAs, miRNAs, circRNAs all impact regulation. Discoveries emerged about control functions, particularly via noncoding RNAs, identified competing, antagonizing, regulating agents biogenesis. On hand, CELFs an intriguing example through broaden our understanding RBP/noncoding regulatory axis. Balancing these complex pathways is undeniably deserves further research. This review outlines some mechanisms regulation functional consequences physiology.

Accumulating evidence has shown that long noncoding RNA (lncRNA) CRNDE functions as an oncogene in many cancer types. However, its clinical value not yet been explored hepatocellular carcinoma (HCC).A total of 166 patients with HCC and 100 healthy volunteers were enrolled this study. The expression levels serum exosomal lncRNA detected controls by quantitative real-time PCR (qRT-PCR).The significantly increased compared normal controls. High was associated tumor size, differentiation, TNM stage. Receiver operating characteristic (ROC) analysis revealed area under the ROC curve (AUC) 0.839, a sensitivity specificity 69.3% 85.0%. In addition, overall survival (OS) disease-free (DFS) longer lower to those higher expression. Moreover, cirrhosis had worse OS DFS than without cirrhosis. Univariate multivariate analyses indicated high independent indicator poor prognosis.Taken together, might serve potential biomarker for diagnosis prognosis.

Accumulating evidence suggests that long noncoding RNAs (lncRNAs) are deregulated in hepatocellular carcinoma (HCC) and play a role the pathogenesis of non-alcoholic fatty liver disease (NAFLD). However, current understanding lncRNAs NAFLD-associated HCC is limited. In this study, transcriptomic profiling analysis three paired human samples from patients with NAFLD-driven adjacent showed LINC01468 expression was significantly upregulated. vitro vivo gain- loss-of-function experiments promotes proliferation cells through lipogenesis. Mechanistically, binds SHIP2 cullin 4 A (CUL4A)-linked ubiquitin degradation, thereby activating PI3K/AKT/mTOR signaling pathway, resulting promotion de novo lipid biosynthesis progression. Importantly, inhibitor reversed sorafenib resistance induced by overexpression. Moreover, ALKBH5-mediated N6-methyladenosine (m6A) modification led to stabilization upregulation RNA. Taken together, findings indicated novel mechanism which LINC01468-mediated lipogenesis progression CUL4A-linked degradation SHIP2. acts as driver NAFLD, highlights potential LINC01468-SHIP2 axis therapeutic target for HCC.
