Abstract Wound healing is an extremely complex process involving multiple levels of cells and tissues. It mainly completed through four stages: haemostasis, inflammation, proliferation, remodelling. When any one these stages impaired, it may lead to delayed or even transformation into chronic refractory wounds. Abstract Mastitis is a common and serious bacterial infection of the mammary gland. Saikosaponin A (SSA) triterpenoid saponin isolated from Bupleurum falcatum that has ability to treat various diseases. However, little known about role SSA in achieving mastitis remission. Here, we found alleviated Staphylococcus aureus ( S. )‐induced by attenuating inflammation maintaining blood‐milk barrier integrity. Furthermore, activated nuclear factor kappa B (NF‐κB) pathway upregulated p‐p65 p‐IκB. also induced ferroptosis gland mice, mainly characterized excessive iron accumulation, mitochondrial morphological changes impaired antioxidant production. ‐induced NF‐κB activation were prevented SSA. Moreover, SAA could upregulate expression SIRT1, Nrf2, HO‐1 GPX4. And inhibitory effects on reversed SIRT1 inhibitor EX‐527. In conclusion, protected through suppressing activating SIRT1/Nrf2 pathway.
