Mitochondria play a crucial role in mediating apoptosis and have emerged as promising therapeutic target for the treatment of cancer. Anaplastic thyroid cancer (ATC) stands as the most formidable form of malignancy, presenting a persistent challenge in clinical management. Recent years have witnessed gradual unveiling intricate genetic underpinnings governing ATC through next-generation sequencing. The emergence this landscape has paved way for exploration targeted therapies and immunotherapies trials. Despite these strides, precise mechanisms pathogenesis identification efficacious treatments demand further investigation. Our comprehensive review stems from an extensive literature search focusing on implications, notably pivotal MAPK PI3K-AKT-mTOR signaling pathways, along with ATC. Moreover, we screen summarize advances challenges current diagnostic approaches ATC, including invasive tissue sampling represented by fine needle aspiration core biopsy, immunohistochemistry, 18F-fluorodeoxyglucose positron emission tomography/computed tomography. We also investigate enormous studies prognosis outline independent prognostic factors future assessment therapy By synthesizing literature, aim to encapsulate evolving oncology, potentially shedding light novel pathogenic avenues therapeutic exploration.
