Abstract Background To enhance Niangya yak's reproductive efficiency and breeding process, this study employed Label-free quantitative proteomics technology to analyze fresh (F) chilled (C) semen samples.Result The analysis revealed an extensive repertoire of 18,338 peptide segments 2,579 proteins within yak semen. Among them, 2,300 were successfully annotated in the database. Notably, a total 300 DEPs identified between C F groups; comprising 129 up-regulated 171 down-regulated DEPs. GO enrichment highlighted that these may be involved lipid metabolism as well protein binding with proteases phosphorylation processes associated sperm cryodamage. KEGG showed 35 mapped pathways related motility, plasma membrane integrity apoptosis.
