Inactivated whole tumor cell-based vaccines (WTVs) are a promising strategy for immunotherapy, but have exhibited limited antitumor effects clinically. Aiming at constructing enhanced WTVs, we developed glycopolymer-engineered WTVs (G-WTVs) using Halo-Tag protein (HTP) fusion technique and reversible addition-fragmentation chain transfer (RAFT) polymerization. In our study, G-WTVs with varying molecular weights of glycopolymers were constructed. Compared to unmodified cells, all effectively induced the polarization macrophages toward M1 phenotype promoted secretion pro-inflammatory cytokines. This immune response was attributed improved interactions between macrophages. Among G-WTVs, medium weight variant demonstrated most pronounced enhancement responses. Notably, administration optimized inhibited growth B16 melanoma in mice. Our findings provide new approach enhance efficacy via cell membrane glycopolymer engineering, offering immunotherapy.

Pancreatic cancer (PCA) is a highly malignant tumor with 5-year survival rate of <10%. It characterized as cold an immunosuppressive microenvironment. Liver dysfunction due to biliary obstruction can affect the inflammation index, indicator immune status. Adjusting indices for liver function may enhance their clinical utility predicting overall (OS) in PCA patients. Resected cases were selected using specific criteria. indicators identified by Spearman's analysis integrated into covariance refine indices, including modified neutrophil-to-lymphocyte ratio (mNLR), platelet-to-lymphocyte (mPLR), lymphocyte-to-monocyte (mLMR), systemic immune-inflammation index (mSII), and C-reactive protein (mCRP). These clinicopathological factors analyzed identify independent OS predictors. A nomogram was developed compared primary inflammation-based model calibration curves, decision curve (DCA), concordance (C-index). direct bilirubin (DBIL), indirect (IBIL), alanine aminotransferase (ALT), aspartate (AST), lactate dehydrogenase (LDH), gamma-glutamyl transferase (GGT), alkaline phosphatase (ALP), albumin (ALB) indices. In patients, higher mNLR, mSII, CA19-9, T stage, N stage associated worse OS, while mLMR or PNI levels correlated better OS. Vascular invasion predicted poor whereas chemotherapy improved The model's surpassed that model. incorporating demonstrated superior utility. recommended prognostic prediction, especially patients obstruction. For advanced staging poorly differentiated tumors, intraoperative margin nanoknife ablation timely postoperative adjuvant are prognosis.
