Summary Coriander ( Coriandrum sativum L. 2 n = x 22), a plant from the Apiaceae family, also called cilantro or Chinese parsley, is globally important crop used as vegetable, spice, fragrance and traditional medicine. Here, we report high‐quality assembly analysis of its genome sequence, anchored to 11 chromosomes, with total length 2118.68 Mb N50 scaffold 160.99 Mb. We found that two whole‐genome duplication events, respectively, dated ~45–52 ~54–61 million years ago, were shared by family after their split lettuce. Unbalanced gene loss expression are observed between duplicated copies produced these events. Gene retention, expression, metabolomics comparative genomic analyses terpene synthase (TPS) involved in terpenoid biosynthesis pathway contributing coriander’s special flavour, revealed tandem contributed coriander TPS expansion, especially compared carrot counterparts. Notably, highly expressed all 4 tissues 3 development stages studied likely major‐effect encoding linalool myrcene synthase. The present sequencing, transcriptome, metabolome efforts provide valuable insights into evolution spice trait biology other related plants, facilitated further research functions improvement.
