Candidalysin, a cytolytic peptide toxin secreted by the human fungal pathogen Candida albicans, is critical for pathogenesis. Yet, its intracellular targets have not been extensively mapped. Here, we performed high-throughput enhanced yeast two-hybrid (HT-eY2H) screen to map interactome of all eight Ece1 peptides with their direct protein and identified list potential interacting proteins, some which were shared between peptides. CCNH, regulatory subunit CDK-activating kinase (CAK) complex involved in DNA damage repair, was as one host candidalysin. Mechanistic studies revealed that candidalysin triggers significantly increased double-strand breaks (DSBs), evidenced formation γ-H2AX foci colocalization CCNH γ-H2AX. Importantly, binds directly activate CAK inhibit repair pathway. Loss alleviates DSBs under treatment. Depletion candidalysin-encoding gene fails induce stimulates upregulation murine model oropharyngeal candidiasis. Collectively, our study reveals acts hijack canonical pathway targeting promote infection.

ABSTRACT Candida albicans is an opportunistic fungal pathogen that can cause life-threatening systemic infections and distressing mucosal infections. A major breakthrough in understanding C. pathogenicity was the discovery of candidalysin, first cytolytic peptide toxin identified a human pathogenic fungus. Secreted by hyphae encoded ECE1 gene, this 31-amino acid integrates into permeabilizes host cell membranes, causing damage across diverse types. Follicle stimulating hormone (FSH) stands as one of the most prevalently used reproductive hormones in field animal-assisted reproduction. Conventionally, pituitary FSH is sourced from heterologous glands pigs and sheep procured slaughterhouses, it typically exists form crude FSH. The specific challenges inherent FSH-based assisted reproduction drugs has significantly spurred interest exploring novel alternatives, aiming to reduce reliance on these traditional sources relevant production processes. In this study, α- β-FSH genes were retrieved cDNA libraries. These selected construct a recombinant protein-the cow (CrFSH)-through application homologous recombination method. Notably, β-subunit was extended by carboxy-terminal peptide (CTP). After successfully integrating two into Chinese hamster ovary (CHO) cells, protein (approximately 33 kDa) culture supernatant detected using Western blotting (WB). results GCs proliferation experiment indicated that both 1.2 µg/mL pFSH 20-20,000 ng/mL CrFSH could promote vitro. Remarkably, 4th day after treatment, 20 had higher rate than μg/mL (p < 0.001).
