Candidalysin, a cytolytic peptide toxin secreted by the human fungal pathogen Candida albicans, is critical for pathogenesis. Yet, its intracellular targets have not been extensively mapped. Here, we performed high-throughput enhanced yeast two-hybrid (HT-eY2H) screen to map interactome of all eight Ece1 peptides with their direct protein and identified list potential interacting proteins, some which were shared between peptides. CCNH, regulatory subunit CDK-activating kinase (CAK) complex involved in DNA damage repair, was as one host candidalysin. Mechanistic studies revealed that candidalysin triggers significantly increased double-strand breaks (DSBs), evidenced formation γ-H2AX foci colocalization CCNH γ-H2AX. Importantly, binds directly activate CAK inhibit repair pathway. Loss alleviates DSBs under treatment. Depletion candidalysin-encoding gene fails induce stimulates upregulation murine model oropharyngeal candidiasis. Collectively, our study reveals acts hijack canonical pathway targeting promote infection.

ABSTRACT A knowledge‐based interactome maps interactions among proteins and molecules within a cell using experimental data, computational predictions, literature mining. These interactomes are vital for understanding cellular functions, pathways, the evolutionary conservation of protein interactions. They reveal how regulate growth, differentiation, development. Transitioning to functionally validated is crucial in developmental biology (Evo‐Devo), especially non‐model species, uncover unique regulatory networks, novelties, reliable gene interaction models. This enhances our complex trait evolution across species. The European Evo‐Devo 2024 conference Helsinki hosted first fish‐specific symposium, highlighting growing interest fish Advances genome annotation, editing, imaging, molecular screening expanding research. High‐throughput data have enabled deduction networks. next steps involve creating species‐specific interactomes, validating them functionally, integrating additional deepen Evo‐Devo. short review aims address logical this transition, as well necessities limitations journey.

SUMMARY The human microbiome, including bacteria, fungi, archaea, and viruses, is intimately linked to both health disease. relationship between bacteria disease has received much attention intensive investigation, while that of the fungal also known as mycobiome, lagged far behind bacteria. There growing evidence showing mycobiome dysbiosis in cancer patients, certain cancer-specific fungi may contribute progression by interacting with host It was demonstrated role fungi-derived products should not be underestimated. Therefore, investigating how pathogenesis contributes onset spread would yield crucial information for diagnosis, prevention, anti-cancer therapy.

ABSTRACT Candida albicans is an opportunistic fungal pathogen that can cause life-threatening systemic infections and distressing mucosal infections. A major breakthrough in understanding C. pathogenicity was the discovery of candidalysin, first cytolytic peptide toxin identified a human pathogenic fungus. Secreted by hyphae encoded ECE1 gene, this 31-amino acid integrates into permeabilizes host cell membranes, causing damage across diverse types. Beyond its activity, candidalysin trigger potent innate immune responses epithelial cells, macrophages, neutrophils. Additionally, plays key role nutrient acquisition during infection. This review explores biology activation, extends discussion to non-candidalysin Ece1p peptides, shedding light on their emerging significance.

Follicle stimulating hormone (FSH) stands as one of the most prevalently used reproductive hormones in field animal-assisted reproduction. Conventionally, pituitary FSH is sourced from heterologous glands pigs and sheep procured slaughterhouses, it typically exists form crude FSH. The specific challenges inherent FSH-based assisted reproduction drugs has significantly spurred interest exploring novel alternatives, aiming to reduce reliance on these traditional sources relevant production processes. In this study, α- β-FSH genes were retrieved cDNA libraries. These selected construct a recombinant protein-the cow (CrFSH)-through application homologous recombination method. Notably, β-subunit was extended by carboxy-terminal peptide (CTP). After successfully integrating two into Chinese hamster ovary (CHO) cells, protein (approximately 33 kDa) culture supernatant detected using Western blotting (WB). results GCs proliferation experiment indicated that both 1.2 µg/mL pFSH 20-20,000 ng/mL CrFSH could promote vitro. Remarkably, 4th day after treatment, 20 had higher rate than μg/mL (p < 0.001). Additionally, cyclic adenosine monophosphate (cAMP) induction assay unequivocally confirmed possesses superior activity compared pFSH. findings underscore holds great potential promising candidate for approaches dairy herds.
