Abstract α‐Synuclein (α‐Syn) amyloidogenesis is considered a promising diagnostic marker and therapeutic target for Parkinson's disease (PD). Simultaneously visualizing mitigating α‐Syn are essential future PD theranostics, yet they continue to pose an insurmountable challenge. This study have herein developed nanobody‐decorated polydiacetylene approach straightforward solution. Grafting α‐Syn61‐95 segment into the third complementary determining region of parent nanobody generates engineered X30 that can bind with prevent its through homotypic interaction. It next use decorate poly(deca‐4,6‐diynedioic acid) (PDDA), ultrastrong alkyne Raman signal (2120 cm −1 ) in cellular silent region, create targeting probe PX30. The binding affinity between be further boosted over 150 times attributed rigidity PDDA backbone multivalent effect. Therefore, PX30 not only enables real‐time visualization high signal‐to‐noise ratio living zebrafish, but also alleviates amyloidogenesis‐mediated damage zebrafish embryos by effectively inhibiting at low stoichiometric concentrations scavenging pathologic reactive oxygen species.

Inducing programmed cell death is an efficient strategy for cancer treatments, and a deep understanding of the molecular mechanisms underlying pathways significance rational design anticancer drugs. Herein, propiolamide-mediated crosstalk between ferroptosis apoptosis investigated. In situ monitoring reactive oxygen species (ROS) formation structural changes propiolamide compounds achieved by Raman spectroscopy. The propiolamides in inducing monooxygenase-mediated ROS generation inhibiting GPX4 activities are revealed. Furthermore, pro-ferroptotic pro-apoptotic roles containing terminal alkynes verified. This study provides label-free enzyme-drug interactions their dynamics. It first attempt to basis through two important enzymes death. paves way designing novel drugs that capable triggering synergistic contribution multiple forms efficacy.
