Introduction Adrenal tumors (ATs) encompass a wide differential diagnosis, necessitating multi-step process for accurate identification. Liquid biopsy emerges as promising non-invasive technique distinguishing between malignant and benign, well hyperfunctioning non-functioning cases. Recent studies have highlighted the potential of microRNAs circulating biomarkers; however, their clinical utility remains underexplored. This study aims to validate diagnostic performance selected microRNAs, (miR-483-5p, miR-210, miR-335 miR-22-3p), identified through microRNA profiling studies, markers malignancy or cortisol hypersecretion in cohort patients with ATs. Methods We collected serum samples from 75 ATs, including 50 cases adrenocortical adenomas (ACA) 25 carcinomas (ACC), along 15 controls. In ACC subgroup, 16 were obtained preoperatively upon detection recurrence (active group), while remaining disease-free long-term follow-up. Among 56 ATs evaluated (50 ACAs 6 ACC), 26 had tumors, 22 exhibited mild autonomous secretion, 8 Cushing syndrome. Quantitative real-time polymerase chain reaction was employed analyze expression. Results Circulating levels miR-483-5p miR-210 significantly elevated active compared both (p<0.001 p=0.004, respectively) controls (p=0.02 p = 0.03, respectively). Notably, higher (p 0.01). MiR-483-5p demonstrated best accuracy ACAs, achieving sensitivity 81.3% specificity 88%, patients, reaching 89%. MiR-22-3p successfully differentiated syndrome those (area under curve=AUC=0.800, 95% CI: 0.653–0.953, p=0.01) (AUC= 0.800, 0.610–0.990, p=0.02). Additionally, miR-22-3p significant correlation traditional tests hypercortisolism. Conclusion supports liquid approach an innovative method diagnosing monitoring offering complementary tool existing methods.

Glycosylation is a crucial post-translational modification that regulates cellular functions and the surrounding microenvironment. The abnormal expression of glycosyltransferases, essential enzymes in this process, has been linked to tumor development. In addition, long noncoding RNAs (lncRNAs) have shown exert multifaceted regulatory influence on gene expression. Nevertheless, research glycosyltransferase-related lncRNAs (GTRLs) endometrial cancer (EC) still unclear. study, we constructed novel prognostic model based GTRLs using univariate Cox regression, Lasso, multivariate stepwise regression analysis. Our findings identified four were significantly associated with prognosis EC patients. Kaplan-Meier (KM) survival analysis revealed notable reduction overall (OS) among high-risk populations. Furthermore, independent assessments, receiver operating characteristic (ROC) curves nomograms, demonstrated risk scores derived from these outperformed other clinical variables within TCGA-UCEC data set predicting patient prognosis. And low-risk cohort exhibited increased immune infiltration decreased purity. Additionally, significant differences mutation profiles observed, burden (TMB) being higher cohort, suggesting potentially better response immunotherapy. Different subgroups also displayed varying sensitivities several chemotherapeutic agents. Finally, model's validity was confirmed through specimens vitro experiments, which AC090617.5 AP001107.9 can proliferation, migratory functions, cisplatin sensitivity EC. Therefore, features effectively predict guide therapeutic decision-making.

A large fraction of the human genome is transcribed in RNA molecules that do not encode for proteins but have a crucial role regulating almost every level gene expression and, thus, define specific phenotype each cell. These non-coding RNAs include well-characterized microRNAs and thousands less-defined longer transcripts, named long RNAs. Both types markedly affect onset progression numerous pathologies, ranging from cancer to vascular neuro-degenerative diseases. In recent years, substantial effort has been made design drugs targeting ncRNAs, promising advancements produced micro-RNA mimics inhibitors. Each ncRNA controls several targets, overall effect its inhibition or overexpression depends on function set genes it regulates. Therefore, selecting most appropriate target, predicting final outcome ncRNA-based therapies, utilize detailed accurate knowledge their functional interactions. this review, I recapitulate principal resources which collect information microRNA lncRNA networks, focusing non-homogeneity data result disparate approaches. highlight identifiers interaction evidence standardization helping user filter integrate derived different databases reliable web regulative relations.

Abstract The expression levels of microRNAs (miRNAs) are strongly linked to cancer progression, making them promising biomarkers for detection. Enzyme‐free signal amplification DNA circuits have facilitated the detection low‐abundance miRNAs. However, these methods may neglect diagnostic value (or weight) different Here, a molecular computing approach with weighted is presented. Polymerase‐mediated strand displacement employed assign weights target miRNAs, reflecting miRNAs’ values, followed by signals using localized catalytic hairpin assembly. This method applied diagnose miRNAs non‐small cell lung (NSCLC). Machine learning used identify NSCLC‐specific and corresponding optimum classification healthy individuals. With output simplified as single channel fluorescence intensity. Cancer tissues ( n = 18) adjacent 10) successfully classified within 2.5 h (sample‐to‐result) an accuracy 92.86%. strategy has potential extend digital multidimensional biomarkers, advancing personalized disease diagnostics in point‐of‐care settings.
