Abstract Mucosal-associated invariant T (MAIT) cells are a unique subset of innate-like lymphocytes that bridge innate and adaptive immunity. Characterized by their semi-invariant cell receptor (TCR) abundant localization in mucosal tissues, MAIT recognize microbial metabolites, primarily derived from the riboflavin biosynthesis pathway, presented major histocompatibility complex (MHC)-related protein 1 (MR1). This interaction, along with co-stimulatory signals, triggers rapid immune responses, including cytokine secretion cytotoxic activity, highlighting importance maintaining homeostasis combating infections. review provides an in-depth overview biology, development, activation pathways, functional diversity, protective roles immunity, contributions to diseases like cancer inflammatory bowel disease (IBD), context-dependent dual functions health pathology. also highlights emerging therapeutic potential immunotherapy. Their TCR specificity, abundance, tissue-homing properties make them ideal candidates for engineering novel therapies, such as chimeric antigen (CAR)-MAIT cells, targeting infections, cancers, autoimmune diseases. Challenges escape, exhaustion, CAR design optimization must be addressed enhance clinical efficacy. In summary, integral function, presents exciting opportunities treatment wide range Further research is essential unlock full these versatile cells.

Non-alcoholic fatty liver disease (NAFLD) includes a range of hepatic manifestations, starting with steatosis and potentially evolving towards non-alcoholic steatohepatitis (NASH), cirrhosis or even hepatocellular carcinoma. NAFLD is major health burden, its incidence increasing worldwide. Although it primarily disturbed metabolism, involves several immune cell-mediated inflammatory processes, particularly when reaching the stage NASH, at which point inflammation becomes integral to progression disease. The cell landscape diverse steady state further evolves during NASH direct consequences for severity. In this Review, we discuss current concepts related role cells in onset NASH. A better understanding mechanisms by contribute pathogenesis should aid design innovative drugs target therapeutic options are limited. Abstract Alcoholic liver disease (ALD) and nonalcoholic fatty (NAFLD) are the two major types of chronic worldwide. Inflammatory processes play key roles in pathogeneses diseases, continuous inflammation promotes progression alcoholic steatohepatitis (ASH) (NASH). Although both ALD NAFLD closely related to inflammation, their respective developmental mechanisms differ some extent. Here, we review multiple immunological therapeutic targets associated with diseases differences ASH NASH. Multiple cell liver, including macrophages, neutrophils, other immune hepatocytes, involved inflammation. In addition, microRNAs (miRNAs), extracellular vesicles (EVs), complement also contribute inflammatory process, as does intertissue crosstalk between intestine, adipose tissue, nervous system. We point out that plays important promoting repair controlling bacterial infections. Understanding complex regulatory process disrupted homeostasis during development may lead improved targeted intervention strategies.

Liver fibrosis, as an excess deposition of extracellular matrix (ECM) components, results from chronic liver injury well persistent activation inflammatory response and fibrogenesis. fibrosis is a major determinant for disease (CLD) progression in the last two decades our understanding on molecular cellular mechanisms underlying fibrogenic CLD has dramatically improved, boosting pre-clinical studies clinical trials designed to find novel therapeutic approaches. From these several critical concepts have emerged, starting reveal complexity pro-fibrotic microenvironment which involves very complex, dynamic interrelated interactions between different hepatic extrahepatic cell populations. This review will offer first recapitulation established pathophysiological basic principles by intentionally focus attention NAFLD/NASH, metabolic-related form with high impact general population emerging leading cause worldwide.
