Dendritic cells (DCs), central participants in the allergic immune response, can capture and present allergens leading to inflammation immunopathogenesis of rhinitis (AR). In addition initiating antigen-specific responses, DCs induce tolerance modulate homeostasis. As a special type DCs, tolerogenic (tolDCs) achieve mainly by suppressing effector T cell responses inducing regulatory (Tregs). TolDCs suppress modulating tolerance, thereby reducing symptoms AR. Multiple vaccine platforms have been employed to develop the nasal SARS-CoV-2 vaccines in preclinical studies, and dominating pipelines are viral vectored as protein-based vaccines. Among them, several vectored-based entered clinical development. Nevertheless, some unsatisfactory results were reported these studies. In face of such urgent situations, it is imperative rapidly next-generation intranasal COVID-19 utilizing other technologies. Nanobased emerged an approach against respiratory infectious diseases. Harnessing power nanotechnology, offer a noninvasive yet potent defense pathogens, including threat COVID-19. The improvements made mucosal delivery technologies based on nanoparticles, lipid polymeric inorganic nanoparticles etc., not only provide stability controlled release but also enhance adhesion, effectively overcoming limitations conventional Hence, this review, we overview evaluation highlight current barriers. Next, modern systems nanoplatforms summarized. challenges application nanoplatform finally discussed.

Antigen-specific T lymphocytes are the central regulators of tolerance versus immune pathology against otherwise innocuous antigens and key targets antigen-specific therapy. Recent advances in understanding cells allergy resulted from improved technologies to directly characterize allergen-specific by multiparameter flow cytometry or single-cell sequencing. This unravelled phenotypically functionally distinct populations, such as Type 2a helper (Th2a), follicular Th (Tfh), regulatory (Treg), 1 (Tr1), cells. Here we will discuss role different Th-cell subsets healthy state, during sensitization development allergy, induction allergen immunotherapy (AIT). To date, mechanisms AIT only causal treatment not completely understood. The analyses ex vivo support concept specific-Th2(a) cell deletion rather than an expansion Tr1 Treg underlying mechanism.

Abstract Background The nature of epitopes on Bet v 1 recognized by natural IgG antibodies birch pollen allergic patients and pollen‐exposed but non‐sensitized subjects has not been studied in detail. Objective To investigate IgE recognition to study the effects 1‐specific 1‐induced basophil activation. Methods Sera from (BPA, n = 76), without allergy (NBPA, 40) non‐allergic individuals (NA, 48) were tested for IgE, as well 4 reactivity folded recombinant 1, two unfolded fragments comprising N‐terminal (F1) C‐terminal half (F2) peptides spanning corresponding sequences apple allergen Mal d ELISA or micro‐array analysis. ability serum inhibit binding rBet 1‐derivatives was assessed competition ELISAs. Furthermore, modulate activation investigated using rat basophilic leukaemia cells expressing human FcεRI which had loaded with BPA patients. Results react almost exclusively conformational whereas IgG, BPA, NBPA NA recognize mainly sequential epitopes. studies show that specific unfolded/sequential is inhibited hence latter seem represent cryptic did correlate therefore does be a result primary sensitization PR10 allergen‐containing food. Natural only poorly could even enhance Conclusion different These findings explain why allergen‐specific do protect against symptoms suggest have clonal origin.
