Abstract Plasmids carrying antibiotic resistance genes (ARG) are the main mechanism of dissemination in Enterobacterales. However, fitness-resistance trade-off may result their elimination. Chromosomal integration ARGs preserves advantage while relieving selective pressure for keeping costly plasmids. In some bacterial lineages, such as carbapenemase producing sequence type ST38 Escherichia coli , most chromosomally integrated. Here we reproduce by experimental evolution mobilisation bla OXA-48 gene from pOXA-48 plasmid into chromosome. We demonstrate that this depends on a plasmid-induced fitness cost, mobile genetic structure embedding ARG and novel antiplasmid system ApsAB actively involved destabilization. show targets high low-copy number combines nuclease/helicase protein Argonaute-like protein. Mobile genetic elements shape microbial gene repertoires and populations. Recent results reveal that many, possibly most, mobile require helpers to transfer between genomes, which we refer as Hitcher Genetic Elements (hitchers or HGEs). They may be a large fraction of pathogenicity resistance genomic islands, whose mechanisms have remained enigmatic for decades. Together with their helper bacterial hosts, hitchers form tripartite networks interactions evolve rapidly within parasitism–mutualism continuum.
