Kidney stone disease (KSD) (alternatively nephrolithiasis or urolithiasis) is a global health care problem that affects people in almost all of developed and developing countries. Its prevalence has been continuously increasing with high recurrence rate after removal. Although effective therapeutic modalities are available, preventive strategies for both new recurrent stones required to reduce physical financial burdens KSD. To prevent kidney formation, its etiology risk factors should be first considered. Low urine output dehydration the common risks types, whereas hypercalciuria, hyperoxaluria, hypocitraturia major calcium stones. In this article, up-to-date knowledge on (nutrition-based mainly) KSD provided. Important roles fluid intake (2.5–3.0 L/d), diuresis (>2.0–2.5 lifestyle habit modifications (for example, maintain normal body mass index, compensation working high-temperature environment, avoid cigarette smoking), dietary management [for sufficient at 1000–1200 mg/d, limit sodium 2 3–5 g/d chloride (NaCl), oxalate-rich foods, vitamin C D supplements, animal proteins 0.8–1.0 g/kg weight/d but increase plant patients uric acid those hyperuricosuria, proportion citrus fruits, consider lime powder supplementation] summarized. Moreover, uses natural bioactive products caffeine, epigallocatechin gallate, diosmin), medications thiazides, alkaline citrate, other alkalinizing agents, allopurinol), bacterial eradication, probiotics also discussed.

The gut microbiota and its homeostasis play a crucial role in human health. However, for some diseases related to the microbiota, current traditional medicines can only relieve symptoms, it is difficult solve root causes or even cause side effects like disturbances microbiota. Increasing clinical studies evidences have demonstrated that probiotics, prebiotics, postbiotics prevent treat various diseases, but currently they be used as dietary supplements rather than medicines, which restricts application of probiotics field medicine. Here, this review analyzes importance health problems systematically summarizes effectiveness mechanisms maintaining treating based on animal models trials. Several studies have linked gut microbiota to human brain activity. This study used Mendelian randomization (MR) investigate the causal relationship between microbes and delirium.MR was select SNPs from large-scale GWAS summary data on 211 taxa delirium. Inverse variance weighting (IVW), weighted median, MR-Egger methods were for statistical analyses. Outliers assessed using leave-one-out method. To avoid horizontal pleiotropy, we performed MR-PRESSO intercept tests. Cochran's Q I2 values IVW assess heterogeneity.IVW suggested that genetic prediction of family Desulfovibrionaceae (1.784 (1.267-2.512), P = 0.001), order Desulfovibrionales (1.501 (1.058-2.128), 0.023), genus Candidatus Soleaferrea (1.322 (1.052-1.659), 0.016) increased risk delirium, but Oxalobacteraceae (0.841 (0.722-0.981), 0.027), genera Holdemania (0.766 (0.620-0.946), 0.013), Ruminococcus gnavus (0.806 (0.661-0.982), 0.033), Eggerthella (0.815 (0.667-0.997), 0.047) reduced delirium.(1) Limited sample size, (2) inability interactions, (3) limited European populations. Our results suggest presence microbial Desulfovibrionaceae, Desulfovibrionales, whereas family, Holdemania, gnavus, decreased However, potential probiotic interventions in prevention perioperative delirium should be emphasized.

Oxalate is an antinutrient present in a wide range of foods, with plant products, especially green leafy vegetables, being the main sources dietary oxalates. This compound has been largely associated hyperoxaluria, kidney stone formation, and, more severe cases, systematic oxalosis. Due to its impact on human health, it extremely important control amount oxalate particularly for patients issues. In this review, summary and discussion current knowledge analysis, extraction conditions, specific features analytical methods, reported occurrence health implications are presented. addition, brief conclusion further perspectives whether high-oxalate foods truly problematic can be seen as threats shown.

Kidney stone, one of the oldest known diseases, has plagued humans for centuries, consistently imposing a heavy burden on patients and healthcare systems worldwide due to their high incidence recurrence rates. Advancements in endoscopy, imaging, genetics, molecular biology bioinformatics have led deeper more comprehensive understanding mechanism behind nephrolithiasis. stone formation is complex, multi-step long-term process involving transformation stone-forming salts from free ions into asymptomatic or symptomatic stones influenced by physical, chemical biological factors. Among various types kidney observed clinical practice, calcareous nephrolithiasis currently most common exhibits intricate mechanism. Extensive research suggests that primarily originates interstitial subepithelial calcified plaques and/or blockages openings collecting ducts. These eventually come contact with urine renal pelvis, serving as nidus crystal subsequent growth. Both pathways share similar mechanisms, such drive abnormal composition, involvement oxidative stress inflammation, an imbalance inhibitors promoters. However, they also possess unique characteristics. Hence, this review aims provide detailed description present recent discoveries regarding processes two distinct birthplaces: interstitium tubule lumen.

Abstract Oxalic acid is among the most abundant organic acids found in different biospheres, including plants, as an end product of metabolism. It forms either soluble or insoluble salts with monovalent divalent cations, respectively. Then, consumption oxalic acid-rich foods human diets, particularly leafy vegetables (e.g., spinach, tea, and rhubarb), affects minerals absorption such calcium. Meanwhile, its high level blood associated many diseases hyperoxaluria systemic oxalosis thus classified potential anti-nutrients. Various factors have affected levels foods, agricultural traits practices. Hence, current review aimed at rediscovering dietary sources, metabolism, various processes employed to reduce content consequently, health harmful effects. Among them are physical/cooking, chemical, fermentation, biotechnological processing. Recent approaches been attempted produce transgenic crops remodeling oxalate degradation. The form seems be better absorbed more than aiding kidney stones formation. Cooking boiling, microwaving, steaming) appears a useful management strategy and, therefore, lowering oxaluria. present provides new perspectives on processing methods lower essential highlighting their advantages any limitations aid improve these nutritional value consumption.

Hyperoxaluria, including primary and secondary hyperoxaluria, is a disorder characterized by increased urinary oxalate excretion could lead to recurrent calcium kidney stones, nephrocalcinosis eventually end stage renal disease. For high dietary (HDOx) or its precursors intake key reason. Recently, accumulated studies highlight the important role of gut microbiota in regulation homeostasis. However, underlying mechanisms involving metabolite disruptions hyperoxaluria remain poorly understood. Here, we investigated therapeutic efficacy fecal transplantation (FMT) sourced from healthy rats fed with standard pellet diet against excretion, damage (CaOx) crystal depositions via using rat models. We observed dose-dependent increases CaOx due accompanied significant reductions diversity shifts Ruminococcaceae_UCG-014 Parasutterella composition. Metabolomic analysis validated these findings, revealing substantial decreases metabolites associated microbial groups. Transplanting microbes effectively reduced HDOx-induced meanwhile restoring populations their metabolites. Furthermore, FMT treatment significantly decrease kidneys via, at least part, upregulating expressions intestinal barrier proteins transporters intestine. The fact displays optimum pH in acidic range represents big limitation most involving processes occurring at neutral pH, where activity stability are remarkably reduced. Here, through bioinformatics-guided protein engineering followed combinatorial mutagenesis analyses thermal stability, we identified double mutant endowed enhanced catalytic efficiency under physiological conditions. obtained engineered form offers potential tool improved intestinal degradation hyperoxaluria patients.
