Abstract In Alzheimer's disease (AD), microglia are activated by mechanical and biochemical cues in the amyloid‐β (Aβ) plaque‐associated microenvironment, causing neuroinflammation. While impact of Aβ stiffness on microglial activation dynamic interplay between inflammation phagocytosis remain unclear. Here, an vitro microenvironment model is built investigated how plaques triggers via PIEZO1 mechanotransduction pathway. Scanning electrochemical microscopy scanning ion conductance employed to situ monitor reactive oxygen species release, membrane permeability, phagocytic activity microglia. It found that drives early activation, forming oxidative‐stressed impairs integrity And antioxidant‐resveratrol effectively improves dysfunction impaired This work reveals complex among cues, neuroinflammation, suggests potential therapeutic strategies targeting AD.
