Multiple sclerosis (MS) is an autoimmune disease mediated by T helper cells, which characterized neuroinflammation, axonal or neuronal loss, demyelination, and astrocytic gliosis. Histone deacetylase inhibitors (HDACis) are noted for their roles in easing inflammatory conditions suppressing the immune response. Panobinostat, HDACi, now being used treating multiple myeloma. Nevertheless, effect of panobinostat on diseases remains largely unclear. Thus, our research endeavored to determine if administration could prevent experimental encephalomyelitis (EAE) mice, one most commonly animal models MS, further explored underlying mechanisms. The EAE mice were generated then administered continuously with at a dosage 30 mg/kg 16 days. results indicated that markedly alleviated clinical symptoms inhibiting demyelination loss oligodendrocytes central nervous system (CNS). Moreover, decreased inflammation activation microglia astrocytes spinal cords mice. Mechanistically, treatment panobinosat significantly suppressed M1 microglial polarization blocking toll-like receptor 2 (TLR2)/myeloid differentiation factor 88 (MyD88)/interferon regulatory 5 (IRF5) pathway. Additionally, inhibited mitochondrial dysfunction reduced oxidative stress In conclusion, findings reveal ameliorates stress-linked neuroinflammation dysfunction.
