Nocturia, defined as waking one or more times per night to urinate, is a prevalent and burdensome condition with few effective treatments. While the primary risk factor for nocturia advanced age, preclinical studies have addressed pathophysiological mechanisms of in older subjects. Here, we develop translational model using aging mice behavioral paradigm that enables circadian assessment voluntary urination group-housed animals. We discovered dampened diurnal regulation urinary behavior aged compared adult controls. Molecular analyses revealed disrupted expression canonical genes mouse kidney bladder tissues. Notably, identified age-related loss mechanosensory ion channel, Piezo 1, suggesting potential mechanism linking disruption altered sensitivity. Our results reveal role dysfunction identify 1 promising therapeutic target chronobiological intervention.
