Abstract Immune cells need to sustain a state of constant alertness over lifetime. Yet, little is known about the regulatory processes that control fluent and fragile balance called homeostasis. Here we demonstrate JAK-STAT signaling, beyond its role in immune responses, major regulator cell We investigated JAK-STAT-mediated transcription chromatin accessibility across 12 mouse models, including knockouts all STAT factors TYK2 kinase. Baseline signaling was detected CD8 + T macrophages unperturbed mice—but abrogated unstimulated deprived their normal tissue context. observed diverse gene-regulatory programs, effects STAT2 IRF9 were independent STAT1. In summary, our large-scale dataset integrative analysis mutant wild-type mice uncovered crucial cells, where it contributes poised epigenetic transcriptional helps prepare these for rapid response stimuli.

Systemic lupus erythematosus (SLE) is a genetically predisposed, female-predominant disease, characterized by multiple organ damage, that in its most severe forms can be life-threatening. The pathogenesis of SLE complex and involves cells both innate adaptive immunity. distinguishing feature the production autoantibodies, with formation immune complexes precipitate at vascular level, causing damage. Although progress understanding has been slower than other rheumatic diseases, new knowledge recently led to development effective targeted therapies, hold out hope for personalized therapy. However, drugs available date are still an adjunct conventional therapy, which known toxic short long term. purpose this review summarize recent advances disease discuss results obtained from use drugs, look future therapies may used absence current standard care or even cure serious systemic autoimmune disease.

Lichen planus is a chronic disease affecting the skin, appendages, and mucous membranes. A cutaneous lichen rare occurring in less than 1% of general population, while oral illness up to five times more prevalent; still, both forms equally impair patient’s quality life. The etiology not entirely understood. Yet, immune-mediated mechanisms have been recognized since environmental factors such as hepatitis virus infection, mechanical trauma, psychological stress, or microbiome changes can trigger genetically susceptible individuals. According current understanding, immunopathogenesis caused by cell-mediated cytotoxicity, particularly cytotoxic T lymphocytes, whose activity further influenced Th1 IL-23/Th-17 axis. However, other immunocytes inflammatory pathways complement these mechanisms. This paper presents comprehensive insight into actual knowledge about planus, with causal genetic being discussed, described, principal effectors its circuits identified.

Interferon-gamma (IFN-γ) has been identified as a crucial factor in determining the responsiveness to immunotherapy. Produced primarily by natural killer (NK) and T cells, IFN-γ promotes activation, maturation, proliferation, cytokine expression, effector function immune while simultaneously inducing antigen presentation, growth arrest, apoptosis tumor cells. However, cells can hijack signaling pathway mount resistance: rather than increasing antigenicity succumbing death, acquire stemness characteristics express immunosuppressive molecules defend against antitumor immunity. In this review, we summarize potential mechanisms of resistance occurring at two critical stages: disrupted signal transduction along IFNG/IFNGR/JAK/STAT pathway, or preferential expression specific interferon-stimulated genes (ISGs). Elucidating molecular through which develop help identify promising therapeutic targets improve immunotherapy, with broad application value conjugation targeted, antibody cellular therapies.
