Immunotherapy, represented by immune checkpoint inhibitors (ICI), is transforming the treatment of cancer. However, only a small percentage patients show response to ICI, and there an unmet need for biomarkers that will identify who are more likely respond immunotherapy. The fundamental basis ICI immunogenicity tumor, which primarily determined tumor antigenicity antigen presentation efficiency. Here, we propose method measure score (TIGS), combines mutational burden (TMB) expression signature processing presenting machinery (APM). In both correlation with pan-cancer objective rates (ORR) clinical prediction individual patients, TIGS consistently showed improved performance compared TMB other known response. Abstract Purpose: Response rates to immune checkpoint blockade (ICB; anti-PD-1/anti-CTLA-4) correlate with the extent of tumor infiltrate, but mechanisms underlying recruitment T cells following therapy are poorly characterized. A greater understanding these processes may see development therapeutic interventions that enhance T-cell and, consequently, improved patient outcomes. We therefore investigated chemokines essential for cell and subsequent efficacy immunotherapies. Experimental Design: The upregulated by dual PD-1/CTLA-4 were assessed using NanoString-based analysis results confirmed at protein level flow cytometry cytometric bead array. Blocking/neutralizing antibodies requirement key chemokines/cytokines effector cells. Results in patients treated inhibitors single-cell RNA-sequencing (RNA-seq) paired survival analyses.
