Cardiovascular
disease

factors.
One
novel
potential
contributor
CVD
might
be
metabolism

Abstract
Blood
lipids
and
metabolites
are
markers
of
current
health
future
disease
risk.
Here,
we
describe
plasma
nuclear
magnetic
resonance
(NMR)
biomarker
data
for
118,461
participants
in
the
UK
Biobank.
The
biomarkers
cover
249
measures
lipoprotein
lipids,
fatty
acids,
small
molecules
such
as
amino
ketones,
glycolysis
metabolites.
We
provide
an
atlas
associations
these
to
prevalence,
incidence,
mortality
over
700
common
diseases
(
nightingalehealth.com/atlas
).
results
reveal
a
plethora
associations,
including
susceptibility
infectious
risk
various
cancers,
joint
disorders,
mental
outcomes,
indicating
that
abundant
circulating
beyond
cardiometabolic
diseases.

Abstract
Risk
stratification
is
critical
for
the
early
identification
of
high-risk
individuals
and
disease
prevention.
Here
we
explored
potential
nuclear
magnetic
resonance
(NMR)
spectroscopy-derived
metabolomic
profiles
to
inform
on
multidisease
risk
beyond
conventional
clinical
predictors
onset
24
common
conditions,
including
metabolic,
vascular,
respiratory,
musculoskeletal
neurological
diseases
cancers.
Specifically,
trained
a
neural
network
learn
disease-specific
states
from
168
circulating
metabolic
markers
measured
in
117,981
participants
with
~1.4
million
person-years
follow-up
UK
Biobank
validated
model

independent
cohorts.

found

Developing
a
single-domain
assay
to
identify
individuals
at
high
risk
of
future
events
is
priority
for
multi-disease
and
mortality
prevention.
By
training
neural
network,
we
developed
disease/mortality-specific
proteomic
score
(ProRS)
based
on
1461
Olink
plasma
proteins
measured
in
52,006
UK
Biobank
participants.
This
integrative
markedly
stratified
the
45
common
conditions,
including
infectious,
hematological,
endocrine,
psychiatric,
neurological,
sensory,
circulatory,
respiratory,
digestive,
cutaneous,
musculoskeletal,
genitourinary
diseases,
cancers,
mortality.
The
discriminations
witnessed
accuracies
achieved
by
ProRS
10
endpoints
(e.g.,
cancer,
dementia,
death),
with
C-indexes
exceeding
0.80.
Notably,
produced
much
better
or
equivalent
predictive
performance
than
established
clinical
indicators
almost
all
endpoints.
Incorporating
predictors
enhanced
power
most
endpoints,
but
this
combination
only
exhibited
limited
improvement
when
compared
alone.
Some
proteins,
e.g.,
GDF15,
important
discriminative
values
various
diseases.
We
also
showed
that
good
observed
could
be
largely
translated
into
practical
utility.
Taken
together,
profiles
may
serve
as
replacement
complex
laboratory
tests
