This report describes the development of a centrifugally controlled microfluidic dynamic solid-phase extraction (dSPE) platform to reliably obtain amplification-ready nucleic acids (NAs) directly from buccal swab cuttings. To our knowledge, this work represents first centrifugal microdevice for comprehensive preparation high-purity NAs raw samples. Direct-from-swab cellular lysis was integrated upstream NA extraction, and automatable laser-controlled on-board microvalving strategies provided strict spatiotemporal fluidic control required practical point-of-need use. Solid-phase manipulation during leveraged application bidirectional rotating magnetic field promote thorough interaction with sample (e.g., capture). We illustrate broad utility technology by establishing downstream compatibility extracted three noteworthy assays, namely, polymerase chain reaction (PCR), reverse transcriptase PCR (RT-qPCR), loop-mediated isothermal amplification (LAMP). The PCR-readiness DNA confirmed generating short tandem repeat (STR) profiles following multiplexed amplification. With no changes assay workflow, viral RNA successfully contrived (spiked) SARS-CoV-2 samples, RT-qPCR.
