Acrylamide (ACR) is a widespread industrial and food contaminant that garnered considerable attention for its carcinogenic, neurotoxic, reproductive toxic effects. The antioxidant effects of Portulaca oleracea seeds extract (POS) fertility-enhancing were inspiring to evaluate the protective potential pinpoint mechanisms molecular targets UPLC-MS fingerprinted POS on ACR-induced testicular toxicity in rats.Male Wistar rats divided into 6 equal groups negative control, ACR model (10 mg/kg b.wt.), at doses (200 400 b.wt.) POS-treated groups. All treatments given by oral dosing every day 60 days.Administration reversed epididymides weight loss with improved semen quality count, ameliorated ACR-decreased lesion scoring, oxidative stress, degeneration, Leydig cell apoptosis dysregulated PCNA Caspase-3 expression dose-dependent manner. It upregulated declined level serum testosterone steroidogenic genes such as CYP11A1 17β3-HSD an obvious histologic improvement testes re-establishment normal spermatogenic series, Sertoli cells.The supplementation may provide effect dysfunction which mediated antioxidant, antiapoptotic modulatory
