Acanthopanax senticosus is a unique wild resource in Northeast China. Its main active ingredient polysaccharide, which has prominent immunomodulatory effect. In this study, the purified polysaccharide component of leaves(ASPS-A1) with strong activity was isolated by column chromatography, and its structure properties were characterized HPGPC, FT-IR. Results showed that ASPS-A1 mainly composed α- 1,4-D-GalA, 1,5-L-Ara, β- 1,4-D-Gal. RT-qPCR experiments RNA-seq analysis used to study immunoregulatory mechanism ASPS-A1. The results could significantly up-regulate levels cytokines iNOS, IL- 1β, 6, TNF-α activated macrophages through MAPK, NF-κB, Toll-like receptor signaling pathways. Inhibitory further confirmed promotes expression TNF-α, 6 via TLR4 receptor, 1β TLR2 receptor. order identify target ASPS-A1, molecular docking conducted. demonstrated bind both TLR2, forming stable complexes cavities on protein surface hydrogen bonding hydrophobic interaction. scores indicated regulate immune response pathways, particularly interaction TLR4. summary, screened most immunoreactive components disclosed furnished research basis for potential application as natural enhancer.

The shaddock peel polysaccharide (SPP) has various biological activities. This explored its antioxidant activity. In vitro tests, the two SPP components showed good DPPH, hydroxyl radical scavenging ability and anti-lipid peroxidation ability. At a concentration of 3.2 mg/mL, abilities polysaccharides were 21.65% 18.96%, respectively. SPP-3 had slightly stronger to scavenge radicals than SPP-2. When reached order polysaccharides' DPPH rates was SPP-2 > SPP-3. Therefore, strong activity could be used in antioxidant-related applications future.
