A quinine-derived thiourea-promoted enantioselective aza-Friedel-Crafts reaction of 3-aminobenzofurans with isatin-derived ketimines is developed, providing a variety 3-benzofuran-3-amino-2-oxindoles bearing quaternary stereocenter good to excellent yields (72-95%) and moderate enantioselectivities (48-97%). The synthetic potential this concise efficient protocol revealed by gram-scale preparation further transformation the adduct an optically pure spirocyclic oxindole.

Abstract A photoredox defluorinative silylation of α-trifluoromethyl arylalkenes for rapid access to useful gem-difluoroalkenes is disclosed. Stable and easily prepared silacarboxylic acids are used as silyl radical precursors in a photocatalytic decarboxylative process. The mild conditions operational simplicity make our method straightforward strategy construct bearing various functional groups, powerful incorporating gem-difluoroalkene moiety into natural products drug molecules.
