Photothermal therapy (PTT) utilizes localized hyperthermia above 50°C generated by nanomaterials upon exposure to near-infrared (NIR) laser light for effective cancer cell eradication. Yet, in high-temperature PTT, tumor cells develop heat stress tolerance due elevated shock protein (HSP) levels, diminishing therapeutic efficacy. Moreover, excessive can trigger inflammatory conditions and promote metastasis. Targeting the glycolytic pathway highly metabolically active offers a promising strategy inducing starvation therapy, capitalizing on their vigorous energy demands amidst rapid proliferation. Here, Initially, we synthesized luminous NIR-II dye, FNF, followed its encapsulation alongside myricetin (My) within amphiphilic polypeptide carriers through self-assembly method. The resulting nanoparticles exhibited excellent imaging capabilities boasted notable photothermal conversion efficiency of 55.58%. Furthermore, Myricetin effectively impeded glucose transport facilitated transporter 1 (GLUT1), curtailing supply cells. This interference disrupted mitochondrial production, leading decreased adenosine triphosphate (ATP) synthesis subsequent downregulation HSP70 expression. By leveraging this approach, which targets HSP expression via GLUT1 inhibition, enhance efficacy PTT while achieving synergistic effect mild starvation.
