The hexapeptide YPVEPF with strong sleep-enhancing effects could be detected in rat brain after a single oral administration as we previously proved. In this study, the mechanism and molecular of targeted stress-induced anxiety mice were first investigated, its key active structure was further explored. results showed that significantly prolong sleep duration improve indexes, including prolonging time spent open arms center. Meanwhile, by increasing level GABA/Glu ratio, 5-HT, dopamine serum regulating anabolism multiple targets, but blocked bicuculline WAY100135. Moreover, simulation stably bind to vital GABAA 5-HT1A receptors due Tyr-Pro-Xaa-Xaa-Pro-, electrostatic van der Waals energy played dominant roles stabilizing conformation. Therefore, displayed anxiolytic GABA-Glu metabolic pathway serotoninergic system depending on distinctive self-folding structures Tyr two Pro repeats.

Chronic stress is a major inducer of anxiety and insomnia. Milk casein has been studied for its stress-relieving effects. We previously prepared hydrolysate (CP) rich in the sleep-enhancing peptide YPVEPF, this study aims to systemically investigate different protective effects CP on dysfunction anxiety/insomnia behavior underlying mechanisms chronically stressed mice. Behavioral results showed that ameliorated stress-induced insomnia more effectively than milk casein, difference amelioration was highly correlated with an increase GABA, 5-HT, GABAA, 5-HT1A receptors, BDNF decrease IL-6 NMDA receptors Furthermore, restored these dysfunctions brain colon by activating HPA response, modulating ERK/CREB-BDNF-TrκB signaling pathway, alleviating inflammation. The abundant YPVEPF (1.20 ± 0.04%) Tyr-based/Trp-containing peptides may be key reasons compared casein. Thus, work revealed main active structures provided novel dietary intervention strategy prevention treatment chronic-stress-induced behaviors.

Insomnia affects one-third of the world’s population; negative effects insomnia are significant, and traditional medications have numerous side cause considerable suffering. This has aroused interest in obtaining sleep-improving substances from foods. study conducted a comprehensive literature review using Web Science PubMed with keywords like “sleep”, “insomnia”, “food”. A subsequent summary revealed that certain foods, including milk, Ziziphus jujuba, Lactuca sativa, ginseng, Schisandra chinensis, Juglans regia, etc., purported to enhance sleep quality by prolonging duration, reducing latency, alleviating anxiety. The mechanisms these foods’ mainly occur via central nervous system, particularly gamma-aminobutyric acid (GABA)ergic 5-hydroxytryptamine (5-HT)ergic systems. Although this supports fact they potential, further research is needed. There also issues such as more limited fewer mechanisms, pharmacokinetic studies, models being involved. These need be addressed future adequately address problem insomnia. It hoped will contribute into foods properties and, future, provide an effective natural alternative for those seeking medication.

Tourette syndrome (TS) is a common chronic neuropsychiatric disorder with prevalence of approximately 1% in children and adolescents. TS characterized by sudden involuntary motor tics along vocal tics. A pathological study on postmortem patients has reported 50-60% reduction striatal gamma-aminobutyric acidergic (GABAergic) interneurons, suggesting role for GABAergic system imbalances tic development. However, the effect exogenous GABA administration alleviation remains unreported. In this study, we aim to investigate therapeutic effects TS-like behaviors Sprague-Dawley rats explore its potential mechanisms, including gut microbiota regulation, oxidative stress mitigation, restoration GABA-glutamate balance, provide insights into pathogenesis alternative treatment strategies. model rat was established through intraperitoneal 3,3-Iminodipropionitrile (150 mg/kg/day), followed (20 mg/kg/day) gavage. 15 minutes behavioral testing (stereotypical behavior head twitching behavior) then conducted. 16S rRNA sequencing identified microbiome changes, LC-MS assessed metabolite changes. The results showed that 4-week alleviated rats. led an increase Acinetobacter other beneficial bacteria. also significantly upregulated metabolites compared group. By correlation analysis intestinal bacteria, statistical Clostridium_sensu_stricto_1 negatively correlated top 20 differential metabolites. Moreover, changes alterations metabolites, gut-brain axis involvement. Exogenous reducing harmful flora modulating metabolism. Despite challenges like low blood-brain barrier permeability dose safety humans, GABA's may be realized prodrug development optimized dosing. These findings are preliminary require further clinical validation.

As a non-nutritive sweetener, aspartame is widely used in everyday life. However, its safety highly controversial, especially effects on neurobehavior. We evaluated the of chronic daily oral administration aspartame-containing drinking water (at doses equivalent to 7-28% FDA-recommended human DIV) memory and rhythm behaviors mice further investigated changes at molecular level brains. Our results demonstrated that exposed exhibited impairment. Disorders hippocampal neurotransmitter metabolism pathological damage may be responsible for aspartame-induced impairment via inhibition BDNF/TrkB pathway. Furthermore, our findings suggested disturbed clock gene expression hypothalamus after exposure led altered rest-activity behavior, this disruption circadian exacerbate This study highlights negative neurobehavioral provides valuable insights into rational safe use.
