Cisplatin (CP), a widely used antineoplastic drug, could induce hepatotoxicity and is also one of the most common reasons for drug-induced liver injury (DILI). Licorice (Chinese name GanCao, GC) commonly herbal drug in traditional Chinese medicine (TCM) that has been shown to treat diseases DILI. CP documented apoptosis through promotion endoplasmic reticulum (ER) stress. However, exact role ER stress pathogenesis CP-induced remains unclear. A rat DILI model was constructed intraperitoneal injection CP, anti-DILI effect GC detected by coefficients, function tests, pathological staining, oxidative indices. Additionally, indices were investigated quantitative real-time PCR (qRT-PCR), Western blotting, immunofluorescence (IF) on toxicity tissues LO2 cells. In group, indicators significantly elevated, lesions more pronounced, reactive oxygen species (ROS) level increased, expression markers, factors, related protein kinase RNA-like kinase/activating transcription factor 4/C/EBP homologous (PERK/ATF4/CHOP) pathway elevated. Treatment with improved function, reduced lesions, decreased ROS. addition, inhibited PERK/ATF4/CHOP pathway, demonstrating anti-CP-induced GC. this study, we verified protective rats clarified its mechanisms apoptosis.
