The authors carried out a comprehensive review of the application peptides known as cell-penetrating (CPPs) in various drug delivery systems (DDS), with prospect achieving novel solutions and ideas to overcome challenges DDS, by making them more able penetrate cells biological membranes. A conceptual search was conducted relevant literature databases (Scopus, PubMed, Web Science, Google Scholar) up 1 April 2025 using keywords such systems, peptides, CPPs, complexes, conjugates, nanoparticles, dendrimers, exosomes, liquid crystalline, liposomes, micelles, nanospheres lipid nanoparticles. studies demonstrate that antimicrobial effect drugs, including curcumin, gentamicin, antifungal drugs like imidazoacridinone derivatives, can be enhanced when they are conjugated or complexed CPPs. CPPs possess positive charges, which make suitable for gene therapy applications facilitating plasmids siRNAs negative charges modern systems. Medicinal formulations containing combination crystals nanostructured carriers (NLCs) increase penetration skin. Additionally, several investigations showed could have impact on pharmacokinetic pharmacodynamic chemotherapy agents, reducing their side effects. significant potential enhancing penetration, bioavailability, targeting, optimization DDS. By computer modeling designing desirable features conducting clinical studies, new methods treating diseases better achieved.
