Lysine acetyltransferase 8 (KAT8) is a member of the MYST family histone acetyltransferases. It catalyzes acetylation H4 at lysine 16 (H4K16ac) and non-histone proteins. Abnormal upregulation or downregulation KAT8 its associated H4K16ac have been observed in malignant tumors, suggesting close association with tumorigenesis progression. Characterized by structural diversity multi-target mechanisms, natural agents increasingly shown to possess significant antitumor activity. This review focuses on KAT8, summarizing molecular mechanisms regulating tumor development catalyzing substrate protein acetylation, which impacts cell proliferation, cycle regulation, apoptosis, DNA damage repair, autophagy. also systematically discusses pharmacological activities small-molecule that target inhibit including compounds, synthetic drugs, non-coding RNAs.
