Rifaximin and rifampicin are good broad-spectrum antimicrobials. The irrational use of antimicrobial drugs in veterinary clinics could threaten public health food safety. It is necessary to develop a reliable detection method the residue for enhancing rational supervision such drugs, reducing slowing down generation bacterial resistance, promoting animal safety human health. So, this study developed an LC-MS/MS rifaximin residues animal-origin foods. residual homogenized test materials were extracted with acetonitrile-dichloromethane solution or acetonitrile presence anhydrous sodium sulfate vitamin C, purified by dispersible solid phase extraction, determined LC-MS/MS, quantified internal standard method. specificity, sensitivity, matrix effect, accuracy, precision investigated edible tissues cattle, swine, chicken. In addition, stability stock working was also investigated. suitable muscle, liver, kidney, fat, milk, eggs chicken, as well fish shrimp. specificity good, analytes not affected different matrices. Both LOD LOQ two 5 μg/kg 10 μg/kg, respectively. results effects each tissue range 80–120%; there no significant effects. average accuracy foodstuffs origin between 80% 120%, below 20% (RSD). proposed showed performance determination, which be employed purification, tissues. pretreatment procedure samples simple feasible. highly specific, stable, reliable, high precision, met requirements quantitative drug residues.
