Abstract Studies have demonstrated the neuroprotective effect of cannabidiol (CBD) and other Cannabis sativa L. derivatives on diseases central nervous system caused by their direct or indirect interaction with endocannabinoid system‐related receptors molecular targets, such as 5‐HT 1A receptor, which is a potential pharmacological target CBD. Interestingly, CBD binding receptor may be suitable for treatment epilepsies, parkinsonian syndromes amyotrophic lateral sclerosis, in serotonergic plays key role. The aim this review was to provide an overview cannabinoid effects neurological disorders, epilepsy, multiple sclerosis Parkinson's diseases, discuss possible mechanism action, highlighting interactions targets phytocannabinoids. has been shown significant therapeutic epilepsy disease, while nabiximols contribute reduction spasticity are frequent option sclerosis. Major depressive disorder (MDD) is one of the most prevalent psychiatric disorders, with an increasing incidence each year and important socioeconomic burden. Although new treatments are continuously being developed, there no effective monitoring method to determine suitability treatment ensure positive outcomes. Therefore, patients often struggle ineffective antidepressants their potential adverse effects, which halts any future progress in managing disorder. Considering microRNAs (miRNAs) as biomarkers for various pathologies evidence modulation several genes involved MDD, this minireview aimed evaluate literature data on impact miRNAs MDD usefulness response. The correlations between expression support existence a common epigenetic mechanism explain differences influencing efficacy MDD.

The body instinctively responds to external stimuli by increasing energy metabolism and initiating immune responses upon receiving stress signals. Corticosterone (CORT), a glucocorticoid (GC) that regulates secretion along the hypothalamic-pituitary-adrenal (HPA) axis, mediates neurotransmission humoral regulation. Due widespread expression of receptors (GR), effects CORT are almost ubiquitous in various tissue cells. The D2 dopamine receptor (DRD2) gene has garnered substantial attention as one of the most extensively studied genes across various neuropsychiatric disorders. Since its initial association with severe alcoholism in 1990, particularly through identification DRD2 Taq A1 allele, numerous international investigations have been conducted to elucidate role different conditions. As February 22, 2024, there are 5485 articles focusing on listed PUBMED. There 120 meta-analyses mixed results. In our opinion, primary cause negative reports regarding polymorphisms is inadequate screening controls, not adequately eliminating many hidden reward deficiency syndrome behaviors. Moreover, pleiotropic effects variants identified neuropsychologic, neurophysiologic, stress response, social defeat, maternal deprivation, and gambling disorder, epigenetic DNA methylation histone post-translational discussed this article. 70 PUBMED for 20 October 19, 2022. For commentary, we did denote and/or methylation; instead, provided a brief summary based behavioral effects. Based fact that Blum Noble characterized allele generalized necessarily specific alcoholism, it now behooves field find ways either use effector moieties edit neuroepigenetic insults or possibly harness idea potentially removing mRNA-reduced expression by inducing “dopamine homeostasis.”

Stress-related mental disorders have become increasingly prevalent, thus endangering health worldwide. Exploring stress-associated brain alterations is vital for understanding the possible neurobiological mechanisms underlying these changes. Based on existing evidence, endogenous cannabinoid system (ECS) plays a significant role in stress response, and disruptions its function are associated with neurobiology of various stress-related disorders. This study primarily focuses investigating impact chronic unpredictable (CUS) expression hippocampal type 1 (CB1) receptors, part ECS, adult male female Wistar rats. Additionally, it explores whether environmental enrichment (EE) initiated during adolescence could mitigate CUS-associated CB1 expression. rats, shortly after weaning, were placed either standard housing (SH) or EE conditions duration 10 weeks. On postnatal day 66, specific subgroups SH animals underwent 4-week CUS protocol. Western blot (WB) analysis was conducted whole hippocampus left hemisphere to assess total protein expression, while immunohistochemistry (IHC) performed right estimate receptors certain areas (i.e., CA1, CA3 dentate gyrus-DG). The WB revealed no statistically differences levels among groups; however, reduced found sub-regions using IHC. Specifically, significantly decreased receptor CA1 DG both sexes, whereas decrease limited males. Interestingly, proved protective against reductions. These findings suggest region sex-specific endocannabinoid response stress, emphasizing positive early experiences protection adolescent adverse later life.
