Abstract Background Long noncoding RNAs (lncRNAs) are integral to a plethora of critical cellular biological processes, including the regulation gene expression, cell differentiation, and development tumors cancers. Predicting relationships between lncRNAs diseases can contribute better understanding pathogenic mechanisms disease provide strong support for advanced treatment methods. Results Therefore, we present an innovative Node-Adaptive Graph Transformer model predicting unknown LncRNA-Disease Associations, named NAGTLDA. First, utilize node-adaptive feature smoothing (NAFS) method learn local information nodes encode structural fusion similarity network using Structural Deep Network Embedding (SDNE). Next, module is used capture potential association nodes. Finally, employ with two multi-headed attention layers learning global-level embedding fusion. structure coding added as inductive bias compensate missing message-passing mechanism in Transformer. NAGTLDA achieved average AUC 0.9531 AUPR 0.9537 significantly higher than state-of-the-art methods 5-fold cross validation. We perform case studies on 4 diseases; 55 out 60 associations have been validated literatures. The results demonstrate enormous graph incorporate uncovering lncRNA-disease correlations. Conclusions Our proposed serve highly efficient computational associations.

Abstract Globally, the incidence and death rates associated with cancer persist in rising, despite considerable advancements therapy. Although some malignancies are manageable by a mix of chemotherapy, surgery, radiation, targeted therapy, most malignant tumors either exhibit poor responsiveness to early identification or endure post-treatment survival. The prognosis for prostate (PCa) is unfavorable since it perilous lethal malignancy. capacity phytochemical nutraceutical chemicals repress oncogenic lncRNAs activate tumor suppressor has garnered significant attention as possible strategy diminish development, proliferation, metastasis, invasion cells. A potential technique treat enhance sensitivity cells existing conventional therapies use phytochemicals anticancer characteristics. Functional studies indicate that modulate drug resistance, stemness, invasion, angiogenesis, proliferation via interactions suppressors oncoproteins. Among them, numerous lncRNAs, such HOTAIR, PlncRNA1, GAS5, MEG3, LincRNA-21, POTEF-AS1, support development PCa through many molecular mechanisms, including modulation regulation various signal pathways like PI3K/Akt, Bax/Caspase 3, P53, MAPK cascade, TGF-β1. Other particular, MALAT-1, CCAT2, DANCR, LncRNA-ATB, ZEB1-AS1, SChLAP1, H19, key players regulating aforementioned processes. Natural substances have shown promising benefits against altering essential signaling pathways. overexpression advanced TNM stage, chemoresistance, reduced LncRNAs possess crucial clinical transitional implications PCa, diagnostic prognostic biomarkers, well medicinal targets. To impede progression beneficial target aberrant long non-coding RNAs using antisense oligonucleotides small interfering (siRNAs). This prevents them from transmitting harmful messages. In summary, several precision medicine approaches may be used rectify dysfunctional lncRNA regulatory circuits, so improving detection eventually facilitating conquest this disease. Due their presence biological fluids tissues, they serve novel biomarkers. Enhancing treatments mitigates resistance chemotherapy radiation.

ABSTRACT Circulating lncRNAs have emerged as promising biomarkers for the diagnosis of various cancers. This study aimed to establish an accurate risk prediction model based on serum facilitate detection prostate cancer (PCA). RT‐qPCR was used analyse levels candidate lncRNAs, and four (NEAT1, ARLNC1, FOXP4‐AS1 DSCAM‐AS1) were identified be differently expressed in from 190 PCA patients, 140 benign controls, 170 healthy controls. A ProsRISK score prostate‐specific antigen (PSA) established training set. ROC analysis validation set revealed that demonstrated more powerful capacity discriminating with AUC 0.926 (95% CI: 0.882–0.970) 0.837 0.770–0.904), which significantly higher than those lncRNA panel or PSA alone (all at p < 0.05). Moreover, showed good diagnostic performance I–II patients compared corresponding AUCs 0.905 0.843–0.968) 0.819 0.732–0.907). Our findings indicated constructed could a reliable stratification great potential clinical use improve precision surveillance PCA.
