Paraneoplastic thrombocytosis is common in ovarian cancer and often correlates with disease recurrence. This study aimed to assess the impact of probiotics on platelet counts patients. Enrolled patients paraneoplastic received oral Bifidobacterium longum JBLC-141 for level modulation. Over a 12-month period, were closely monitored recurrence rates. The revealed significant differences between probiotic placebo groups various parameters, including counts, coagulation functions, inflammatory factors, tumor markers, gut microbiota composition. However, no distinctions observed during postoperative stabilization. Oral effectively mitigated count surges improved functions thrombocytosis. Positive effects inflammation, marker levels, composition also noted. Further clinical validation needed confirm probiotics' potential extending stabilization period.

Immune cell therapy based on chimeric antigen receptor (CAR) technology platform has been greatly developed. The types of CAR immune have expanded from T cells to innate such as NK and macrophages, the diseases treated hematological malignancies non-tumor fields infectious autoimmune diseases. Among them, CAR-T CAR-NK observed examples rapid remission in approved clinical trials, but efficacy is unstable plagued by tumor resistance. ABSTRACT Chimeric antigen receptor (CAR) T-cell therapy has revolutionized the treatment of hematological malignancies but been clinically less effective in solid tumors. Engineering macrophages with CARs emerged as a promising approach to overcome some challenges faced by CAR-T cells due macrophage’s ability easily infiltrate tumors, phagocytose their targets, and reprogram immune response. We engineered CAR-macrophages (CAR-Ms) target chondroitin sulfate proteoglycan 4 (CSPG4), an expressed melanoma, several other CSPG4-targeting CAR-Ms exhibited specific phagocytosis CSPG4-expressing melanoma cells. Combining CD47 blocking antibodies synergistically enhanced CAR-M-mediated effectively inhibited spheroid growth 3D. Furthermore, tumor mouse models. These results suggest that CAR-M immunotherapy is for treating melanoma. STATEMENT OF SIGNIFICANCE alternative treatment. targeting CSPG4, phagocytosed vivo . Thus, may be strategy treat patients

Abstract Macrophages often pervade solid tumors, but their nearest neighbor organization is understudied and potentially enables key functions such as phagocytosis. Here, we observe dynamic macrophage clusters in tumors under conditions that maximize cancer cell phagocytosis use reductionist approaches to uncover pathways cluster formation roles for tumor-intrusive pseudopodia, which term ‘intrudopodia’. Macrophage form over hours on low- adhesion substrates after M1 polarization with interferons, including T cell-derived cytokines, yet prove fluid timescales of minutes. Clusters also sort from M2 macrophages disperse the same substrates. upregulate specific cell-cell receptors suppress actomyosin contractility, while both contribute formation, decreased cortical tension was predicted unleash pseudopodia. neighbors tumor spheroids indeed extend intrudopodia between adjacent junctions – at least when are maximized, coordinated help detach individualize cells rapid engulfment. thereby provide a cooperative advantage overcome cohesion.
