Systemic mirtazapine (MRT) delivery for the treatment of pruritis exhibits severe side effects which needs to be addressed. For this purpose, topical nanostructured lipid carriers (NLCs) containing MRT were developed minimize and enhance therapeutic efficacy. The microemulsion method was utilized preparation loaded NLCs final optimized formulation in gel effective application. terms particle size (PS), zeta potential (ZP), polydispersity index (PDI), percentage entrapment efficiency (% EE) by keeping view quantity drug, tween 80 lipids ratio. Optimized nano exhibited a PS 186.3 ± 1.2 nm, with 0.217 0.03 PDI, ZP -26.0 0.2 mV %EE 86.3 0.3%. Fourier transform infrared spectroscopy (FTIR) differential scanning calorimetry (DSC) analysis confirmed compatibility components encapsulation drug inside matrix respectively. Further, gel-based dispersion analyzed rheology textural characterization. prepared hydrogel (MRT gel) had transparent appearance, non-gritty texture, pH, spreadability 322.33 0.25% release 81% 24 hours followed Korsmeyer- Peppas model. Ex vivo skin permeation depicted only 6.20 µg/cm2 across after hours. Skin irritation study showed no signs erythema edema treated group. formulated successfully may used as promising vehicle pruritus.
