This study evaluates the chemical and enzymatic stability of naringin (NRG), identifies its degradation metabolites, assesses in vitro cytotoxicity, validates a high-performance liquid chromatography (HPLC) method for precise quantification. NRG, flavonoid with antioxidant, anti-inflammatory, anticancer properties, faces clinical limitations due to poor solubility, rapid degradation, low bioavailability. While research efforts on this promising compound have largely focused overcoming these through formulation strategies, it is equally necessary complementary focus preformulation studies enhance NRG's therapeutic potential. These represent fundamental step drug development, providing key insights into physicochemical biological properties NRG serving as basis rational design safe effective formulations future research. was analyzed under various temperature pH conditions. Cytotoxicity evaluated 3T3 cells, an HPLC developed validated quantify primary metabolite, naringenin (NRGN). remained stable up 100 °C physiological (1.2, 5.8, 7.4) but degraded at extreme pH, forming NRGN. assays showed toxicity ≤1 mM (viability >80%), whereas 5 significantly reduced viability.
