This study aimed to investigate the content of major bioactive compounds and characterize volatile sensory profiles Coffea canephora flowers their infusions. Dried from six selected genotypes C. trees infusions were analyzed for using HPLC–DAD, while organic (VOC) GC–MS. Terpenoids are valuable chemicals used across industries. Methanol, a nonsugar-based feedstock, offers an eco-friendly approach to terpenoid production. In this study, hybrid methylotrophic pathway was engineered in Serratia marcescens (S. marcescens) HBQA7, which is tolerant both methanol and compounds. The utilizes bacterial dehydrogenase (Mdh) yeast dihydroxyacetone synthase (Das) produce monoterpenes sesquiterpenes from xylose. 13C labeling experiments identified the optimal enzyme pair: Mdh Acinetobacter gerneri Das Pichia angusta, achieving 7.63% enrichment of central metabolic intermediate pyruvate. Deletion pentose phosphate genes (rpiAB) enhanced utilization, 22.99% enrichment. Optimization mevalonate (MVA) biosynthetic enabled production 5.12 g/L shake flask culture Further construction haloarchaea-type MVA geraniol (574.12 mg/L) (-)-α-bisabolol (1256.41 cultures. This study demonstrates first conversion into terpenoids S. marcescens.

: The study of functional compounds is great importance for the production nutrient-rich and flavorful beer. Streptomyces sp. MJM3502 is a promising producer of rufomycins, which are class potent anti-tuberculosis lead compounds. Although the structure, activity, and mechanism main rufomycin 4/6 its analogs have been extensively studied, significant gap remains in our understanding genome sequence biosynthetic pathway MJM3502, metabolic engineering has not yet reported. This study established genetic manipulation platform for strain. Using CRISPR/Cas9-based technology to in-frame insert strong kasO∗p promoter upstream rufB rufS genes BGC, we increased production by 4.1-fold 2.8-fold, respectively. Furthermore, designing recombinant strains inserting encoding cytochrome P450 enzymes led new derivatives. These findings provide basis enhancing valuable natural compounds offer insights into generation novel active products via synthetic biology engineering.
