Abstract Purpose: Resistance to chemotherapeutics is an eminent cause which leads for search of options that help in diminution Pancreatic Ductal Adenocarcinoma (PDAC) by overcoming resistance issues. Caffeic acid (CFA), a polyphenol occurring many dietary foods known show anti-diabetic and anticancer properties potential. Methods: To unveil effect CFA on PDAC we carried out this research cells, following checked combination pre-sensitization effects CFA. Multitudinous web-based approaches were applied identifying targets then getting their interconnections. Results conclusions: Subsequently, manifested affects in-vitro analysis showing IC 50 concentrations 37.37µM 15.06µM Panc-1 Mia-PaCa-2 respectively. Combination index with different drugs was explored gave no significant results leading further investigate pre-sensitizing effects. reduced concentration doxorubicin both cell lines also triggered ROS generation determined DCFH-DA assay. Gene expression revealed acts differently triggers distinct signalling overcome resistance. Collectively, study investigated role as therapeutics mechanism mitigating sensitizing chemotherapeutics.
