The blood-brain barrier (BBB) plays a vital role in protecting the central nervous system (CNS) by preventing entry of harmful pathogens from bloodstream. However, this also presents significant obstacle when it comes to delivering drugs for treatment neurodegenerative diseases and brain cancer. Recent breakthroughs nanotechnology have paved way creation wide range nanoparticles (NPs) that can serve as carriers diagnosis therapy. Regarding their promising properties, organic NPs potential be used effective drug delivery across BBB based on recent advancements. These remarkable ability penetrate using various mechanisms. This review offers comprehensive examination intricate structure distinct properties BBB, emphasizing its crucial function preserving balance regulating transport ions molecules. disruption conditions such stroke, Alzheimer's disease, Parkinson's disease highlights importance developing creative approaches drugs. Through encapsulation therapeutic molecules precise targeting processes vasculature, NP formulations present hopeful strategy improve BBB. We explore changes pathological investigate factors affect successful into brain. In addition, we most systems associated with shown positive results treating ischemic disorders. opens up new possibilities nanotechnology-based therapies cerebral diseases.

Abstract Medicines, food packaging, personal care products, and cosmetics extensively use polyethylene terephthalate nanoplastics (PET-NaPs). However, they also have harmful impacts on several organs. Betaine demonstrates potent antioxidant anti-inflammatory characteristics. Our goal was to investigate the detrimental impact of PET-NaPs mouse brain evaluate neuroprotective properties betaine. We allocated 40 completely mature male Swiss albino mice into four distinct groups: control group, betaine betaine-co-treated group. Following a 30-day duration, euthanasia performed mice, analyzed tissue samples were obtained from cerebrum, cerebellum, hippocampus. resulted in an elevated level malondialdehyde upregulated cyclooxygenase-2 interleukin-1 beta (IL-1β) expression while significantly reducing levels glutathione downregulating acetylcholinesterase. The PET-NPs caused significant changes histopathology tissue, there demonstrable rise immunostaining IL-1β glial fibrillary acidic proteins. Consequently, effectively alleviated negative consequences PET-NaPs. Therefore, possesses capacity mitigate neurotoxic induced by

Background: Oxidative stress is a key therapeutic target in neurological disorders. As processing wastes from the peanut industry, skins are great sources of antioxidants and possess potential neuroprotection. Methods: We prepared skin extract (PSE) investigated its protective effects against tert-butyl hydroperoxide (t-BHP)-induced oxidative injury HT-22 neuronal cells. Results: PSE was rich phenolic compounds (123.90 ± 0.46 mg GAE/g), comprising flavonoids (75.97 0.23 RE/g) proanthocyanidins (53.34 1.58 PE/g), displayed potent radical scavenging activities chemical-based assays. In cells, pretreatment restored balance endogenous antioxidant defense disrupted by t-BHP, as evidenced significant reductions ROS generation lipid peroxidation levels, along with enhanced antioxidants. Specifically, 25 μg/mL reduced levels 53.03%, decreased MDA content 78.82%, superoxide dismutase (SOD) activity 12.42%, improved ratio glutathione (GSH) to oxidized (GSSG) 80.34% compared t-BHP group. Furthermore, rescued mitochondrial membrane collapse, inhibited cytochrome c (Cyt.c) release, prevented subsequent apoptotic death. Notably, neuroprotective efficacy comparable that edaravone, an approved drug. Mechanistic investigations combining network pharmacology experimental validation revealed PI3K/Akt/Nrf2 signaling pathway played pivotal role mediating PSE. Compared t-BHP-treated µg/mL significantly upregulated PI3K/Akt phosphorylation, expression Nrf2, downstream proteins heme oxygenase-1 (HO-1) NAD(P)H dehydrogenase quinone 1 (NQO1). Conclusions: Collectively, these findings demonstrate natural agent oxidative-related

Background Edaravone dexborneol (Eda-Dex), a promising neuroprotectant composed of edaravone and (+)-borneol, has been clinically applied in stroke treatment. However, the mechanism action Eda-Dex remains unclear. Methods A rat model cerebral ischemia/reperfusion injury (CIRI) was created through middle artery occlusion. Neurological scoring, TTC staining, laser speckle imaging were used to assess neurological deficits, infarct size blood flow (CBF). Behavioral tests, including open field test, elevated plus maze, novel object recognition conducted animal behavior. Western blotting ELISA employed levels expression components NRF2/ARE NF-κB/AIM2 pathways specific cytokines. The oxidative stress markers analyzed via commercially available kits. HE Nissl immunohistochemistry pathological alterations brain. Results dramatically reduced deficit score size, increased CBF, attenuated anxiety-like behavior improved cognitive function CIRI rats. significantly relieved inflammatory response it upregulated NRF2, NQO1, HO-1, SLC7A11 downregulated NF-κB, AIM2, ASC caspase 1 infarcted Moreover, clearly damage, rescued neurons, activation microglia astrocytes. Conclusions results this study confirm that exerts neuroprotective effects by synergistically inhibiting inflammation
