HIF-1α plays a crucial regulatory role in vascular calcification (VC), primarily influencing the osteogenic differentiation of VSMCs through oxygen-sensing mechanisms. Under hypoxic conditions, stability increases, avoiding PHD and VHL protein-mediated degradation, which promotes its accumulation cells then activates gene expressions related to calcification. Additionally, modulates metabolic state by regulating pathways that govern switch between glycolysis oxidative phosphorylation, thereby further advancing process. The interaction other signaling pathways, such as nuclear factor-κB, Notch, Wnt/β-catenin, creates complex network serves critical driving force VC. Therefore, deeper understanding mechanism during development progression VC is great significance, it not only key molecular marker for pathological mechanisms but also represents promising target future anti-calcification therapies.
