ABSTRACT Powassan virus (POWV) is an emergent tick-borne flavivirus that causes lethal encephalitic disease and chronic neurologic deficits in surviving patients. POWV-LI9 a tick-derived isolate neurovirulent age-dependent lethality mice. Serial passage of VeroE6 cells infected with LI9 resulted eight amino acid changes POWV strain LI9P. LI9P fails to cause neurological sequelae, or C57BL/6 mice yet elicits neutralizing antibody responses protects from challenge. Analysis revealed LI9, but not LI9P, present at high levels the CNS, suggesting restricted neuroinvasion CNS replication. are distinguished by D308N envelope change within domain associated cell attachment. Dengue, West Nile and Zika viruses are vector-borne flaviviruses responsible for numerous disease outbreaks in both Hemispheres. Despite relatively low mortality, infection may lead to potentially severe situations such as (depending on the virus): hypovolemic shock, encephalitis, acute flaccid paralysis, Guillain-Barré syndrome, congenital malformations (e.g., microcephaly) and, some situations, death. Moreover, also have major socioeconomic repercussions, especially already vulnerable societies. Thus far, only generic symptoms relief is possible, there no specific treatments available yet.
