Introduction Inactivated vaccines were delivered to low- and middle-income countries during the early pandemics of COVID-19. Currently, more than 10 inactivated COVID-19 have been developed. Most contain an whole-cell index SARS-CoV-2 strain that is adjuvant. Whole virions with aluminum hydroxide among most commonly used. However, emerging variants waning immunity two doses after 3 months, WHO many local governments recommended booster-dose program especially heterologous platform vaccine.

Heterologous prime-boost has broken the protective immune response bottleneck of COVID-19 vaccines. however, underlying mechanisms have not been fully elucidated. Here, we investigated antibody responses and explored germinal center (GC) to priming with inactivated vaccines boosting heterologous adenoviral-vectored or homologous in mice. Antibody were dramatically enhanced by both regimens. immunization induced more robust GC activation, characterized increased Tfh cell populations helper function. Additionally, B-cell activation production observed a regimen. Libra-seq was used compare differences S1-, S2- NTD-specific B cells between vaccination, respectively. S2-specific CD19+ presented somatic hypermutations (SHMs), which mainly enriched plasma cells. Moreover, booster dose promoted clonal expansion specific S2 NTD regions. In conclusion, functional role following SARS-CoV-2 vaccination may be important for modulating responses. These findings provide new insights development that induce response.

Abstract People living with HIV (PLWH) are particularly vulnerable to SARS‐CoV‐2. This multicentre prospective cohort study evaluated the long‐term immunogenicity and safety of a third homologous dose Sinovac CoronaVac in PLWH China. A total 228 127 HIV‐negative controls were finally included followed up for 6 months. Fewer participants reported mild or moderate adverse reactions, no serious events observed. The median levels neutralizing antibodies (nAbs) immunoglobulin G against receptor‐binding domain spike protein (S‐IgG) (655.92 IU/mL, IQR: 175.76–1663.55; 206.83 85.20–397.82) comparable those control group (1067.16 239.85–1670.83; 261.70 77.13–400.75), reached their peak at 4 weeks, exhibiting delayed pattern compared 2‐week group. After then, immune titres gradually decreased over time, but most still maintained positive seroconversion 6‐month mark. Multivariable generalized estimating equation analysis indicated that CD4+T cell count, viral load, antiretroviral therapy (ART) independent factors strongly associated response (each p < 0.05). We suggested should maintain well‐controlled status through ART receive timely administration second booster optimal protection.

ABSTRACT Vaccination against the wild-type (WT) severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) virus did not produce detectable levels of neutralizing antibodies (NAbs) BA.5 strain before it emerged. However, disease-2019 (COVID-19) severity varied highly between unvaccinated, partially vaccinated, and fully vaccinated individuals, for unknown reasons. We assessed infection risk XBB reinfection measured serum NAbs WT, BA.5, XBB.1.9.1 SARS-CoV-2 strains at varying time points in 1,373 individuals who received zero, one, two, or three WT vaccine doses. found that two to doses significantly increased NAb reduced incidence COVID-19-associated pneumonia upon compared zero one dose. Regarding reinfection, those were infected with variant exhibited a lower RNA analysis revealed differentially expressed genes dose unvaccinated groups enriched B cell activation, cytokine-cytokine receptor interaction, complement, monocyte activation functions—indicating vaccination antibody response inflammation. Our results suggest multiple antigen exposures either matched unmatched SARS-COV-2 variants, through infection, may be necessary achieve significant immune imprinting. IMPORTANCE The administration vaccines do perfectly match viral become has been impact resultant illness severity—although precise mechanism underlying this phenomenon remains unclear. clearance, as well inflammatory cytokines wild-type, variants among such strain-mismatched vaccines. Notably, ≥2 COVID-19 appeared stimulate production specific during new attenuate enhance clearance. Such regimens can also reduce reinfection. These findings important guiding development future strategies target both mismatched variants.

The COVID-19 vaccination program has probably been the most complex and extensive project in history until now, which a challenge for all people involved planning management of this program. Patients with immune-mediated inflammatory diseases (IMIDs) on immunosuppressive therapy have required special attention, not only because particular haste carrying out process but also uncertainty regarding their response to vaccines. We now strong scientific evidence that supports hypothesis inhibits humoral vaccines against other infectious agents, such as influenza, pneumococcus hepatitis B. This led same could happen vaccine. However, fact withholding or interrupting when dealing remains unclear. On basis, our article tries compile information available effect immunosuppressant agents responses patients IMIDs proposes an algorithm these patients.

Background: COVID-19 became a global health crisis in early 2020, and the way out of was rapid development vaccines by Sinopharm, Pfizer, Sputnik, among others, which played crucial role controlling pandemic. Therefore, this study aims to investigate long-term immune response measuring levels anti-S1 IgG antibodies induced homologous heterologous vaccination regimens. Methods: We investigated titer antibody produced for viral surface antigen 3, 6 months after second dose, before third 1, dose. Results: Anti-S1 significantly increased three/six dose following booster individuals without prior infection who received all three vaccine doses. The group that initially responded poorly Sinopharm showed significant sustained increase receiving Pfizer booster. Additionally, SARS-CoV-2 between primary boosted titers individuals, regardless used. highest efficacy observed during period declined over time, especially omicron-dominant period. Conclusions: results suggest while doses can enhance levels, type administered influence duration magnitude response.
