Thymus-committed regulatory T cells (Tregs) are essential for immune homeostasis. Recent findings stress their heterogeneity, suggesting possible alternate routes thymic Treg development with unique features in humans, namely the clear evidence of commitment at double-positive (DP) stage and presence a significant population CD8 single-positive (SP) FOXP3 pos Tregs. Here, we present dedicated analysis strategy to spectral flow cytometry-based study thymus from children aged adults (≥ 74-years-old), further elucidate heterogeneity human thymus. We applied an unsupervised pipeline data generated 6 high-dimensional panels, taking advantage common backbone 11 markers, were able map thymocytes along cell maturation stages. Generating UMAP FlowSOM cluster coordinates backbone, projected all other markers onto these, characterizing clusters information markers. Focusing this on events inside putative total gate, could portray rarer subsets Tregs investigate trajectories using pseudotime analysis. uncover within DP uniquely expressing or CD25, DP-branching trajectory towards CD103 CD8SP endpoint, define CD4SP Tregs, including CXCR3 that may consist resident recirculating do not expand elderly. Our cytometry approach separates populations likely distinct functions facilitates design future studies unravel complexity cells.

Apart from rare cases such as lymphomas, germ cell tumors, neuroendocrine neoplasms, and thymic hyperplasia, mass lesions (TMLs) are typically categorized into cysts, thymomas. However, the classification results cannot be determined in advance can only confirmed through postoperative pathology. Therefore, objective of this study is to rely on clinical parameters radiomic features extracted chest computed tomography (CT) scans facilitate preoperative TMLs. The model development specifically focused cysts thymomas, these most commonly encountered anterior mediastinal tumors practice. This retrospective included 400 participants 3 hospitals between September 2017 2024 due were classified 7 groups based ultimately etiology: including types A, AB, B1, B2, B3, C. All underwent contrast-enhanced CT scans, with senior radiologists delineating regions interest extract features. Additionally, participants' ages also collected for analysis. randomly allocated a training set validation at 7:3 ratio. A classifier models developed using data set, their performances evaluated set. exhibited good performance accuracies 0.8547. assist early diagnosis personalized treatment strategies patients
