Glucose-regulated protein 94 (Grp94/gp96) is endoplasmic reticulum (ER) resident form of the 90 kDa heat shock (Hsp90) that responsible for folding, maturation and stabilization more than 400 client proteins. Grp94 has been implicated various diseases including metastatic cancer, primary open-angle glaucoma, infectious diseases. In fact, plays critical roles in different stages viral infection cycle. It chaperones receptor proteins glycoproteins are necessary entry replication. Beyond its role homeostasis, modulates host cellular processes such as apoptosis immune responses, which often exploited by viruses to sustain infection. This work provides an overview pathogenesis across involvement modulation with development Grp94-selective inhibitors their potential anti-viral therapeutics.
